OBJECTIVE To explore the effects of Tea polyphenols and paclitaxel for inhibiting esophageal cancer Eca-109cell growth,inducing apoptosis and mechanism involvedMETHODS The log phase HL-60cells were collected,and divided into TP-group, Paclitaxel-group, TP+Paclitaxel-group,and control group. Cell survival was observed by MTT assay, morphologic changes were viewed under light microscope and cell cycle and cell apoptosis were overviewed by flow cytometry,and the expression of Caspase-3mRNA by RT-PCR (Reverse Transcription-Polymerase Chain Reaction)RESULTS Cell proliferation inhibition rate of Eca-109cells in different-concentration Tp or Paclitaxel was different, and related to concentration,time. The inhibition was more remarkably in TP+Paclitax groups than in separate groupf Compared with the control group,Tea polyphenols induced G1phase improved remarkably;but Paclitaxel,alone or in combination for24h,induced G2/M phase of Eca-109cells,improved remarkably. The TP+Paclitaxel groups changed more remarkably than the seperate group. Compared with the seperate group, the Caspase-3mRNA expression was improved than the Eca-109cells treated with the combination groups for24h.CONCLUDIONS Both TP and Paclitaxel could inhibit growth and induce apoptosis of Eca-109cells,TP+Paclitaxl groups were more obvious,the molecular mechanism may be combined with the increasement of Caspase-3mRNA expression.
