Objective:Small RNA molecules, called microRNAs, freely circulate in human plasma and correlate with varying pathologies. The present study was to investigate the expression of circulating microRNA-1(miR-1) and microRNA-133(miR-133) in viral myocarditis (VM) in children, and to analyse the relationship between the expression changes of miRNAs and the extent of myocardial lesions in VM. With the study, we expect to explored microRNAs’s diagnostic and therapeutic potential in viral myocarditis in children.Methods:40cases of children with viral myocarditis from2012.04-2013.08in Qi lu hospital of Shandong University and Tengzhou People’s Central Hospital were studied. The40patients were divided into two groups:severe cases group (n=20) and mild cases group (n=20), according to the results of the left ventricular ejection fraction (LVEF) and cardiac troponin I examined by echocardiography and enzyme linked immunosorbent assay. All the blood samples were obtained at acute phase and recovery phase, At the same time,20cases were in convalescent stage, who need regular revisited, were selected as the control.Result:①The expression levels of miR-1and miR-133significantly decreased in acute stage and recovery stage? especially in the severe cases group,there was statistical difference comparing with those of in control group (P<0.01);②There is a strong negative correlation between the expression of miR-1,miR-133and the content of CK-MB and cTn-I in the peripheral blood;③There is favorable positive correlation between the expression of miR-1、miR-133and the date of left ventricular ejection (LVEF) and fraction shortening (FS) in acute period, but not in recovery period.Conclusion:The expression and regulation of miR-1and miR-133were abnormal in children with viral myocarditis,which may results in malignant arrhythmias and heart dysfunction in VM. Detection the expression levels of circulating miR-1and miR-133may provide important diagnostic value, and the mircroRNAs intervention may become a new therapeutic method for treatment of viral myocarditis in the near future.
