ObjectiveTo observe the effect and its mechanism on invasion and metastasis to human hepatoma of compatibility of Gallic acid and Curcumol,which are respectively from Rhizoma Curcumae and Euphorbia Helioscopia in ALC.And then to explain the substance for ALC of detoxification and antiliver cancer via eradicating blood stasis and phlegm.Those would offer clinical ideas and theoretical basis for the development of new anti-tumor medicine.MethodsHuman hepatoma cell SMMC-7721were cultured in vitro.We took Cisplatin as a positive control group, while the experimental groups consisted of different concentrations of Curcumol, Gallic acid and the combination of both. After treated with groups of experimental drugs,MTT assay was used to detect the effects on cell proliferation;apoptotic fluorescence intensity and morphology were observed by Hoechst33258staining kit;MTT assay, scratch method and Transwell chamber were taken to evaluate ability of adhesion, migration, invasion for the cells; Real time-PCR and Western Blot assay were applied to assess the regulations on expression of some genes and proteins interrelated invasion and metastasis.Results1.Curcumol and Gallic acid effectively could inhibit the growth of human hepatoma cells SMMC-7721in a time-and-concentration-dependent;2.The fluorescence intensity of the nucleus from experimental drugs groups was significantly higher than the blank one in dose-dependent,and typical apoptotic nucleus can be seen under a microscope;3.The ability of adhesion, migration, invasion for the cells were obvious suppressed after dealed with medications,which were positively correlated with the dose;4. There were downregulation of MMP2、microRNA-21and upregulation of TIMP2、RECK in drug-treated groups,whose extent depended on the concentration;5.Compatibility of Curcumol and Gallic acid has a more significant effect than the same concentration of monomerins in the inhibition on proliferation, invasion and metastasis behavior,promotion on apoptosis,and regulation on expression of some genes and proteins interrelated invasion and metastasisof human hepatoma cells SMMC-7721.ConclusionCompatibility of Curcumol and Gallic acid may be an effective combination in ALC,which could significantly inhibit the invasion and metastasis behavior of hepatocellular carcinoma cell.Downregulation the expression of MMP2、microRNA-21and upregulation of TIMP2、RECK might be one of the mechanisms.
