| Objective: Apply Ion torrent sequencing technology to detect EGFR mutation ofNSCLC,and do statistical analysis about EGFR mutation rate and mutation types, toexplore the clinical applicability of Ion torrent sequencing technology.Methods: Seventy-four cases of non-small cell lung cancer tissue samples andforty-seven cases of normal lung tissue samples were collected, then applied Ion torrentsequencing technology to detect EGFR gene mutation of NSCLC, and do statisticalanalysis for various types of mutations of EGFR gene exon18~21.Results: A total of74non-small cell lung cancer patients were enrolled.43(58.1%)females and31(41.9%) males, between28~80years old, the average age was60.51±11.23years. There were38cases older than61years, accounted for51.4%,36casesaged youngse than61years old, accounting for48.6%. The most frequent type wasadenocarcinoma, a total of51cases (68.9%), squamous carcinoma (19cases) accountingfor25.7%and4cases (5.4%) other types of non-small cell lung cancer.24cases (32.4%)have smoking history,50(67.7%) patients are never-smoker. There were17cases(23.0%) Tumor Node Metastasis phase â… (â… A+B),13cases (17.6%)phase â…¡ (â…¡ A+B),34cases (45.9%) phase â…¢ (â…¢ A+B)and10cases (13.5%) phase â…£. The EGFRexon18~21gene mutations were detected by Ion torrent sequencing technology inseventy-four cases of NSCLC tissue samples, the mutation of exon18was missensemutation,which mainly occurred in the base sequence719th codon; The mutations ofexon19were all the bases deletion mutations,which mainly occurred in the746~753codons,the mose frequent type was del E746-A750.Exon20mutations occurred mainlyin the base sequence768th,768th,774th codons, they were all the bases missensemutations; Exon21mutations located in base sequence848th,858th,861th codons,they were all the missense mutations, especially L858R. The mutation rate of EGFRgene was about41.9%(31/74); There were12cases that exon19mutated, accounting for38.7%; Exon21mutated in16cases, accounting for51.6%, the mutation rate wasabout90.3%in exon19and exon21. Exon20mutated in3cases, accounting for9.7%,exon18mutated in2cases, accounting for6.5%. There were significant differencesabout EGFR gene mutation rate in gender, smoking status and the comparison amongpathological types (P<0.5); there was no significant difference in the lung cancerstaging and age,(P>0.5). The EGFR gene types of normal lung tissue were all wildtypes.Conclusions:The EGFR mutation rate in non-small cell lung cancer cases is about41.9%, including about90.3%in exon19and exon21. |
