Objective:To observe the effect of TUBEX modification PRP on the adipogenicdifferentiation and proliferation of rabbit adipose—derived stem cells.Methods:By the method of enzymatic digestion to obtain rabbit adipose—derived stemcells, and induce them to differentiate into different types cells. Platelet-rich plasmawas prepared by TUBEX modification and traditional centrifugal method. Theadipogenic differentiation abi1ity of the different groups were evaluated using oil RedO staining and the ability of proliferation was detected using CCK8.Results:1under an inverted microscope, morphological meet stem cell morphology,after14days,oil red O staining and alizarin red staining, were successful coloringrespectively.2By the count, platelet of Tubex group was significantly higher than thetraditional preparation.3compared to normal induction group, more cells of the two group wer stained,extracted oil red O content is higher,the group of Tubex preparation is higher than thetraditional preparation group.4compared to normal induction group, more cells of the two group wer alived,the count of OD is higher,the group of Tubex preparation is higher than the traditionalpreparation group.Conclusion: the cells was identified as mesenchymal stem cells by the morphology andnumber of induction of differentiation in vitro。The dengsity of platelet of Tubexmodification is higher than traditional centrifugal method. The group interfusion withPRP by Tubex odification caused a significant increase in dipogenic differentiation ofthe cells.
