The papillary thyroid carcinoma(PTC) is a common one of the head and neck malignant tumors, but the mechanism of its pathogenesis and lymph node metastasis is still unclear.Occurrence and development are complex processes and a variety of genes and their protein products involved in these processes. Recent years studies suggest that mismatch repair system is closely related to the occurrence and development of tumor. But different tumor types vary mismatch repair system changes.hMLH1and hMSH2are two important members of mismatch repair system, and the relationships between mismatch repair genes and papillary thyroid carcinoma is rarely reported; PCNA is not only a factor reflecting the proliferation, but also a key factor participating in mismatch repair. The present experiment detected expressions of hMLH1、hMSH2and PCNA protein by immunohistochemistry in40cases of papillary thyroid carcinoma tissues and28cases normal thyroid tissues paracancerous, at the same time, detected expressions of hMLHl and hMSH2mRNA by in situ hybridization in40cases of papillary thyroid carcinoma tissues and28cases normal thyroid tissues paracancerous.All the dates were analyzed by SPSS16.0statistical software package.The Chi-was used for the comparison of positive rates;the Spearman’s correlation analysis was used for analyzing the correlation of two variables.The level of significant difference is p<0.05.The aim is to investigate the relationship between the expressions of hMLH1, hMSH2, PCNA and the occurrence and development and metastasis of papillary thyroid carcinoma, and to provide theoretical basis for further research on papillary thyroid carcinoma of the biological characteristics and for predicting the prognosis of the carcinoma.Experimental results show:1. The positive expression rates of hMLH1protein and hMLHl mRNA in thyroid papillary cancer were:27.5%(11/40),22.5%(9/40), the positive expression rates in normal thyroid tissue paracancerous were:82.1%(23/28),78.6%(22/28), the positive expression rate of hMLH1protein and hMLH1mRNA in papillary thyroid carcinoma was significantly lower than in adjacent normal thyroid tissue (P <0.05); The positive expression rates of hMSH2protein and hMSH2mRNA in papillary thyroid carcinoma were:82.5%(33/40),85.0%(34/40), the positive expression rates in adjacent normal thyroid tissue were:21.4(6/28),14.3%(4/28), the positive expression rate of hMSH2protein and hMSH2mRNA in papillary thyroid carcinoma was significantly higher than in adjacent normal thyroid tissue (P<0.05); The positive expression rate of PCNA protein in papillary thyroid carcinoma tissue and adjacent normal tissue were:85.0%(34/40),10.7%(3/28), the positive expression rate of PCNA protein in papillary thyroid carcinoma was significantly higher than in adjacent normal thyroid tissue (P<0.05).2. The expressions of HMLH1protein and hMLH1mRNA aren’t related to thyroid papillary patients’ age, gender, tumor diameter size (P>0.05), are closely correlated with tumor envelope invasion and lymph node metastasis(P<0.05); The expressions of hMSH2protein and hMSH2mRNA are closely correlated with papillary thyroid carcinoma of the lymph node metastasis (P<0.05), have no obvious correlation with patients’ age, gender, tumor diameter size and tumor envelope invasion (P>0.05); The expression of PCNA protein isn’t related to thyroid papillary patients’ age, gender, tumor diameter size (P>0.05), is closely correlated with tumor envelope invasion and lymph node metastasis (P<0.05)3. In papillary thyroid carcinoma tissue, the expressions of hMSH2and PCNA protein were positively correlated (P<0.05), the expressions of hMLHl protein and hMLH1mRNA were positively correlated (P<0.05), the expressions of of hMSH2protein and hMSH2mRNA were positively correlated (P<0.05)The results show that the positive expression rate differences of hMLH1protein and hMLH1mRNA, hMSH2protein and of hMSH2mRNA, PCNA protein between adjacent normal tissue and papillary thyroid carcinoma tissue were statistically significant, the abnormal expression of hMLHl, hMSH2and PCNA may prompt the genesis and development of papillary thyroid carcinoma. The findings also show that the expressions of hMLH1, hMSH2and PCNA protein had no correlation with patient gender, age and tumor size, they were all linked to lymph node metastasis, the abnormal expressions of hMLHl and PCNA protein were closely related to tumor capsule invasion; The expressions of hMLHl mRNA and hMSH2mRNA had no correlation with correlated with sex, age, tumor size, but were closely related to lymph node metastasis, the abnormal expression of hMLHl mRNA is also related to the tumor capsule invasion. This experiment show that hMSH2protein and PCNA protein overexpressed in papillary thyroid carcinoma, both were positively related, suggesting hMSH2and PCNA may be in a synergistic effect in the development of papillary thyroid carcinoma and lymph node metastasis; The expressions of hMLHl and hMSH2gene and protein in papillary thyroid carcinoma were in remarkable corelationship, suggesting that hMLH1and hMSH2gene and protein expression in papillary thyroid carcinoma have consistency.
