Object:A(3-induced apoptosis play an important role in the pathogenesis of Alzheimer’s disease. To investigate the effect of Icariin on Aβ25-35induced cell apoptosis in cultured rat pheochromocytoma cells (PC12cells). To determine the mechanisms of Icariin on Aβ-induced apoptosis, we investigated the role of PI3K/Akt pathway in the effect of icariin on Aβ-induced apoptosis.Methods:(1) Effects of Aβ25-35(10μmol/L、20μmol/L、30μmol/L、40μmol/L) on the cell viability and cell apoptosis of PC12cells were determined by MTT assay and flow cytometry assay respectively.(2) Effects of Icariin (2.5μmol/L、5.0μmol/L、10.0μmol/L.20.0μmol/L) on Aβ induced PC12cell viability and apoptosis were determined by MTT assay and flow cytometry assay respectively.(3) PC12cells were randomly divided into five groups:control group, Aβ group, Icariin group, LY294002(Inhibitor of PI3K/Akt pathway) group, Icariin+LY294002group. MTT assay and flow cytometry assay were used to measure the cell viability and apoptosis of PC12cells. Western-blotting assay was used to measure the expression of Akt、p-Akt、bcl-2、bax、caspase-3.Results:(1) Aβ could decrease the cell viability of PC12cells in a dose-dependent manner. Aβ could induce the apoptosis of PC12cells in a dose-dependent manner.(2) Icariin could decrease the apoptosis ratio and increase the cell viability of PC12cells induced by Aβ in a dose-dependent manner.(3) The expression of p-Akt/Akt、bcl-2decreased significantly in the PC12cells treated by Aβ. The expression of caspase-3、bax decreased significantly in the PC12cells treated by Aβ. Icariin could inhibit the increase of caspase-3、bax and decrease of p-Akt/Akt、bcl-2induced by Aβ. LY294002-inhibitor of PI3K/Akt pathway-could inhibit the decrease of cell apoptosis、caspase-3、bax and increase of cell viability、p-Akt/Akt、bcl-2induced by icariin.Conclusion:Icariin could inhibit Aβ-induced cell apoptosis by regulate the expression of caspase-3、bax、bcl-2via the activation of PI3K/Akt pathway.
