Objiective:In our study, we successfully used high fat diet and high fat and high salt diet to feed beagle dogs about three months to established the hypertensive model dogs.Then,we unplement percutaneous catheter-based approach by using radiofrequency energy to disrupt renal sympathetic nerves to verify its antihypertensive efficacy.Furthermore, we measured the plasma concentration of S-100B and NSE to evaluate the relationship between S-100B and NSE with the renal nerve injury.Methods:Beagles (n=16) were assigned into high-fat diet group (n=6), high-fat and high-salt diet group (n=6) and control group (n=4). After baseline measurements, Beagles were fed with lard oil and sodium supplemented diet for three months.After three months, Beagles were assigned into the surgery group (n=8) and the Sham-operated group (n=4).They were fed less fat diet for maintaining the body weight.Pre-operative, and post-operative blood pressure were measured by non-invasive tail cuff sphygmomanometer while the plasma concentration of NSE and S-100B were measured by ELISA.Results:1) Beagles in high-fat diet and high-fat high-salt diet groups gained significant weight and had significant higher levels of the mean arterial pressure compared to its baseline and control groups (P<0.05). However, there was no significant change in heart rate and plasma sodium concentration (P>0.05). And the levels of the blood pressure achieved the diagnostic criterion for hypertension. There was no significant changes of arterial pressure, heart rate and plasma sodium in control group (P>0.05).2) Post-operative systolic blood pressure, diastolic blood pressure, and mean arterial pressure in surgery group were significantly lower compared to sham-operated group on1month and3months after operation (P<0.05). There is no significant difference of heart rate, body weight and plasma sodium between two groups(P>0.05). Three months after surgery, renal angiography revealed no sign of renal artery stenosis.3) Post-operative nervous system injury factor, such as S-100B and NSE expression in surgery group were higher than those of sham-operated group and control group. The plasma concentration of S-100B or NSE had shown significant change at3day,l week,2week and1month after operation(P<0.05).Conclusions:1) Renal sympathetic denervation could significantly reduce the systolic blood pressure, diastolic blood pressure, and mean arterial pressure in hypertensive dogs.2) The plasma concentration of S-100B or NSE can be used as indicator for assessment of renal nerve injury after Renal Sympathetic Denervation.
