Objective:Our group has found that human gastric mucosa epithelial cells were cultured with different H.pylori strains would lead to the gene of TRAF1expression differently, and we have found that the expression of TRAF1differently in chronic gastritis, atrophic gastritis and gastric cancer tissues through immunohistochemical method. Therefore, we further study the gene of TRAF1expression in different gastric mucosa disease which infected H.pylori, and analyze the relationship between TRAF1and virulence genotypes of H.pylori.Methods:First, we collect tissue of different gastric mucosa disease which infected H.pylori. Then, we extract RNA and protein from the tissue, and detect the expression of TRAFl through the Real-Time PCR and Western Blot respectively. Meanwhile we extract DNA of H.pylori which attached to the tissue, and clear the genotypes of H.pylori through the PCR. And according to the the TRAF1expression whether raised we divided them into the TRAF1upregulation group and no upregulation group. Analyse virulence genotypes of H.pylori between the two groups and relationship between TRAF1and virulence genotypes of H.pylori.Results:1. The mRNA expression of TRAF1relative to GAPDH were differently in different gastric mucosa disease.There are significant differences between gastritis and gastric cancer group, atrophic gastritis and gastric cancer group (P<0.05).The protein expression of TRAF1were differently in four groups. Compared with gastric cancer, the expression of gastritis and atrophic gastritis had significant difference(P<0.05).In addition, gastritis and intestinal metaplasia with atypical hyperplasia group had significant difference(P<0.05). There was no significant difference between the rest of two groups.2. The TRAF1express upregulation group of71cases, no upregulation group of63cases.The positive rate of cagA,vacAsl,vacAs2,vacAml,vacAm2,iceAl and iceA2were76.1%,74.6%,20.1%,72.4%,21.6%,52.2%,37.3%respectively. Among them,cagA,vacAsl and vacAml had significant difference between the TRAF1upregulation group and no upregulation group. The genotype of cagA/vacAs1/m1H. pylori strains may strong proliferation of gastric mucosal cells, make the TRAF1expression enhanced, result in the occurrence of gastric cancer.Conclusions:The expression of TARF1were differently in four groups, it is up-regulated in gastric cancer group and intestinal metaplasia with atypical hyperplasia group, this may prompt TRAF1play a important role in the process of the development of gastric cancer. The genotypes of cagA,vacAsl and vacAml had significant difference between the TRAF1upregulation group and no upregulation group. The genotype of cagA/vacAs1/m1H. pylori strains may strong proliferation of gastric mucosal cells, make the TRAF1expression enhanced, result in the occurrence of gastric cancer.
