Objective:To observe the change of miRNAs expression between APPswe/PS△E9double transgenic mice and the matched mice, and to explore the possible effect of miRNAs on the pathogenesis of Alzheimer disease.Methods:We chose6-month-old APPswe/PS△E9double transgenic mice as an experimental group, and6-month-old wild mice of the same species C57as a control group, We examined the miRNAs expression in the brain tissue by miRNAs gene chip and the results were comfirmated by real-time PCR.Results:In miRNAs gene chip analysis identified twelve pieces of miRNA were either up-regulated or down-regulated more than twice in the APPswe/PS△E9double transgenic mice, among them3up-regulated and9downregulated. Compared with the matched group, the The5pieces of miRNA downregulated have statistically significance(P<0.05), which are miR-135a-5p, miR-135a-2-3p, miR-298-5p, miR-466b-3p and miR-669f-3p. Real-time PCR further verify the relative expression levels of the above5miRNAs in the experimental group and the control group were0.73±0.27and1.08±0.58;2.47±1.15and1.65±0.67;0.72±0.14and1.31±0.73;0.57±0.34and1.06±0.35;0.63±0.26and1.02±0.18. Statistical significance was shown in miR-135a-5p, miR-298-5p, miR-466b-3p and miR-669f-3p (P<0.05).Conclusion:MiR-135a-5p, miR-298-5p, miR-466b-3p and miR-669f-3p in the brain of APPswe/PS△E9double transgenic mice are expressed abnormality, which may play an important role in the pathogenesis of Alzheimer disease.There are totally6figures,7tables and55references. |
