Objective:To detect the expression of NGB and HIF-1a in gliomas of different clinical features, histological type and WHO degree and the correlation with Ki-67proliferation index, analysis the relationship and mechanism between the expression of NGB and HIF-la in gliomas, expecting to provide a guide for diagnosis, treatment and prognosis of gliomas.Methods:98pathologically diagnosed gliomas were collected at the Xiangya Hospital and the Third Xiangya Hospital of Central South University from March2011to March2012. I detected the expression of NGB, HIF-la and Ki-67in gliomas by an immunohistochemical method. Kruskal-Wallis rank sum test was used for analysis the expressionof NGB and HIF-la in gilomas of different clinical features, histological type and WHO degree while spearsman correlation test was used for analysis the correlation between each pair of the expression of NGB, Ki-67and HIF-1a in gliomas by SPSS13.0software.Results:1. The total positive rate of NGB and HIF-1a in the98gliomas are88.76%and71.43%respectively. The percentages of NGB express negative, weak positive, moderate positive, strong positive respectively are11.22%,33.67%,27.55%and16.33%while he percentages of HIF-la express negative, weak positive, moderate positive, strong positive respectively are28.57%,38.78%,24.49%and8.16%.98gliomas showed different proliferation activity.40.82%showed weak positive of Ki-67expression. Only7.14%showed strong positve of Ki-67expression.2. Positive expression rate of NGB in gliomas of different age, gender and sympotom groups has no signification difference.(P>0.05).3. Positive expression rate of HIF-la in gliomas of different age, gender and sympotom groups has no signification difference.(P>0.05).4. In gliomas of four different histological types, such as astrocytomas, oligodendroglioma, oligoastrocytic tumors and ependymoma, the expression level of NGB was significantly different respectively. There was significant difference of NGB expression between every two different WHO levels, too. The higher the WHO degree is, the stronger the NGB expressed (P<0.05).5. In gliomas of four different histological types, such as astrocytomas, oligodendroglioma, oligoastrocytic tumors and ependymoma, the expression level of HIF-1a was significantly different respectively. There was significant difference of HIF-1a expression between every two different WHO levels, too. The higher the WHO degree is, the stronger the HIF-1a expressed (P<0.05).6. The higher the WHO degree is, the stronger the Ki-67expressed (P<0.05). Both NGB and HIF-la were positive correlated with Ki-67positive index through spearsman correlation analysis method (NGB rs=0.47and HIF-1a rs=0.48, P<0.05).7. NGB was positive correlated with HIF-1a positive index through spearsman correlation analysis method (rs=0.60, P<0.05).Conclusions:1. The expression levels of NGB and HIF-1a in gliomas of different histological types are significantly different. It indicates that NGB and HIF-1a are helpful for the differential diagnosis in them.2. Both NGB and HIF-1a have the correlation with WHO degree and Ki-67proliferation index in gliomas, testing such expression may in accordance with malignancy evaluation and prognosis of gliomas.3. Analysis of the relationship and mechanism between the expression of NGB and HIF-la in gliomas, it is showed that NGB’s mechanism have significant correlation to HIF-la. The research of the mechanism of these two may provide a new starting point to the treatment of gliomas. Additional evidences are needed to support this viewpoint.
