Objective:To detect the function of dendritic cells in the peripheral blood of HBeAg-positive versus HBeAg-negative patients, thus provide new evidence for the treatment of chronic hepatitis B (CHB) patients. Methods:10CHB patients with HBeAg-positive and10CHB patients with HBeAg-negative were recruited to each experimental group,10heathy persons as control.30ml peripheral blood was collected from each participant under aseptic conditions, which added with heparin to prevent coagulation. Mononuclear cells were isolated from peripheral blood by lymphocyte separation medium density grade centrifugation and transformed to DCs by GM-CSF,IL-4. The morphological changes of cells were observed under the converted microscope, and the level of IL-12secreted by DCs was detected by enzyme linked immunosorbent assay (ELISA) and Reverse transcription PCR. The phenotypes of CD86and CD14of DCs were measured by flow cytometry.Results:The mature DCs in each group had the typical dendritic structure, while their expression of CD14was down-regulated. The level of IL-12and CD86in DCs in the experimental group were significantly lower than that in the control group and significantl higher in HBeAg-positive than that in HBeAg-negative group (all P<0.05).Conclusion:The function of DCs of the CHB patients was deficient, which was better in HBeAg-positive patients than that in HBeAg-negative patients.
