| ObjectiveTo observe expression changes of Janus kinase2(JAK2), signaltransducers and activator of transcription3(STAT3) in the renal interstitial ofIgA nephropathy model,and explore protection mechanism of tripterygiumwilfordii through JAK2/STAT3signaling pathway of IgA nephropathy renaltubular interstitial injury after the application of tripterygium wilfordii.Methods81male SD rats were randomly divided into control group, model groupand treated group, each group has27. Model group and treated groupbuilded IgA nephropathy model, treated group was given tripterygium wilfordii,9rats were put to death in4,9,15weeks of each group,24h urea werecollected for urinary protein detection and we took carotid artery blood fortesting BUN and SCr before they dead; we observed kidney tissuesmorphology, used immunohistochemical method and western blot to detectJAK2and STAT3of renal interstitial.Results1. Immunofluorescence results show that when building model finished at9th week model group and treatment group can be seen obviously fluorescentmaterial deposition at glomerular mesangial area; control group had nofluorescent material deposition;Then it proved that build model were success. 2.Comparing with control group, model group and treated group had asignificantly increased of24hUpro, BUN, SCr; After giving tripterygiumwilfordii, compared to model group, they fell down in treated group(P<0.05).3.Renal histopathologic morphology of control group were normal, therewere glomerular mesangial cell and matrix proliferation,glomerular mesangialarea expanded,capillary extrusion;the renal tubular epithelial cells swelleddegenerated, interstitial inflammatory cell infiltrated.4.JAK2and STAT3expression of model group and treated group in renalinterstitium were significantly higher than those in control group (P <0.01), andafter using tripterygium wilfordii,their expression of treatment group wassignificantly lower than model group (P <0.05).ConclusionsThe protein expression of JAK2and STAT3significantly increase in renalinterstitial of IgA nephropathy model, and both of them decreased obviouslyafter application of tripterygium wilfordii, JAK/STAT signaling pathway may beplay an important role in in development and incidence of renal tubularinterstitial injury IgA nephropathy, tripterygium wilfordii might achieveprotection to renal interstitial of IgA nephropathy through the signal pathway. |
