ObjectiveTo observe the effect of curcumin in the lung tissue of pulmonary fibrosisrats the Bcl-xl、Bax、casepase-3and its role in the treatment of pulmonaryfibrosis.Methods40SD rats, weighing180±10g, were randomly divided into normal controlgroup (A), model group (B), prednisone intervention group (C), and curcumingroup (D)。There are10rats in each group.Lung fibrosis model was bulit byintratracheal injection of bleomycin (5mg/kg) in B, C, D group. All groups aftermodeling was gavage administration the next day, acetate-prednisonesuspension (0.56mg/kg) in group C, curcumin suspension(200mg/kg) in Dgroup, saline in the blank control group and model group).The rats were deadafter weighing in the experiment28d and reserved the lung tissue formorphological observation and tissue homogenates.1. Preparing HE staining,to observe changes of lung tissue pathology morphological by lightmicroscopy;2. Detecting Bcl-xl, Bax expression changes of apoptosis relatedgenes in the lung tissue by RT-PCR method;3. detecting Bcl-xl, Bax, caspase- 3mediated protein content changes of apoptosis-related genes in lung tissueby WB method.Results1.The general situation of pulmonary fibrosis in rats,A group getsustained growth; B group slow weight gain; C, D group rats weight sustainedgrowth, but relatively slow growth of A group,28d in B group compared with Agroup, the difference was statistically significant (P <0.01); C, D groupcompared with B group, the difference was statistically significant (P <0.05).2.The effect of curcumin on rat lungs and lung coefficient: A group oflung surface was pink, smooth surface, good elasticity; B group of lung is pale,the volume decreases, and a hard, nodules, pulmonary change funicular fiberbar visible surface; C, D group lungs appearance pathological changes weresignificantly better than in group B, Lungs show dark pink, not seen noduleformation on the surface, the texture is soft and elastic. A group rat lungcoefficient is5.87±0.28,B group10.23±1.02,6.34±0.33group C7.12±0.41,Dgroup7.98±0.33.B group rat lung coefficient increased significantly, with therest of the group differences were statistically significant (P <0.05).3.The process of pulmonary fibrosis lung tissue structural changes: Agroup does not appear pathological morphological. Alveolar and alveolar wallstructures were normal; B group pathological structural changes, the alveolarwall thickening, incomplete part of the alveolar wall rupture, pulmonary septalthickening, microscope, fibroblasts, cells were cord-like distribution, pulmonaryfibrosis scar exist, some capillaries visible clogging; C, D group rat alveolarwalls slightly thicker than in A group, no fracture of the alveolar walls, mirrors,not seen under the pulmonary interstitial fibrosis scarring.4.Curcumin in lung tissue Bcl-xl, Bax, Caspase-3protein expression:thedetection of Bcl-xl protein Western blotting method, compared with A group, B group significantly increases the expression of Bcl-xl、Bax、casepase-3proteinin lung tissue of rats, the difference was statistically significant difference(P<0.01);Compared with B group, C、D group significantly decreases theexpression of Bcl-xl、Bax、casepase-3protein in lung tissue of rats, thedifference was statistically significant difference (P <0.05);Group C comparedwith group D, there was no statistically significant difference (P>0.05).5.The effect of curcumin on rat lung tissue Bcl-XL, Bax mRNAexpression:the detection of Bcl-xl mRNA RT-PCR method, compared with Agroup, B group significantly increases the expression of Bcl-xl、Bax mRNA inlung tissue of rats, the difference was statistically significant difference (P<0.01);Compared with B group, C、 D group significantly decreases theexpression of Bcl-xl、Bax mRNA in lung tissue of rats, the difference wasstatistically significant difference (P<0.05);Group C compared with group D,there was no statistically significant difference (P>0.05).Conclusions1.In bleomycin-induced pulmonary fibrosis in rat lung tissue, Bcl-xl, Bax,caspase-3gene and protein are present abnormal expression of thephenomenon, the formation of pulmonary fibrosis disease may be abnormalexpression of these apoptosis-related factors.2.Curcumin has therapeutic effect of pulmonary fibrosis in rats, candelay the progression of the disease, improve the quality of life of rats, Thecurcumin may reduce Bax/Bcl-xl expression, decreased caspase-3proteinexpression, reducing the abnormal apoptotic activity in the lung tissue,delaying disease progression of pulmonary fibrosis.3.Curcumin in the anti-apoptosis of pulmonary fibrosis with prednisoneacetate tablets on the treatment effect of similar curative effect.
