Hyperlipidemia, atherosclerosis, and coronary heart disease are the most harmful to public health. The main pathogenic factor, especially the high level of cholesterol, is the disorder of lipid metabolism. There has been an upsurge in finding ways to lower plasma cholesterol levels in such people.The effects of four prebiotic oligosaccharides that polydextrose, polyxylose, inulin, and arabinogalactan(AG) on the level of blood lipid in hyperlipidaemia hamsters were explored, and their mechanism of action was analyzed. Healthy male Syrian hamsters were randomly divided into6groups (n=9). The animal model of hyperlipidemia was established by feeding diets enriched in high cholesterol, in the meantime the four prebiotics were given at a level of5%in the diets for6weeks. The level of serum lipid (total cholesterol(TC), triglycerides(TG), high density lipoprotein cholesterol(HDL-C)), the level of liver lipid (TC,TG), and the content of short chain fatty acid (SCFA, acetic acid, propionic acid, butyric acid) in feces of hamsters were tested. Then the effects of the prebiotics on gene expression of HMG-CoA-R and LDL-R in the liver of hamsters were analyzed.The results were as follow:1. The levels of TC, non-HDL-C and TG in serum of hamsters fed on a lipid rich diet obviously were increased, which showed that hamsters were hyper-responsive to the increasing cholesterol.2. The four prebiotics could reduce the levels of TC, TG and MDA in serm and liver tissue of hamsters significantly (p<0.05,0.01), while the level of TG and HDL-C in serum did not changed in all groups during the experiment.3. The four pieces of prebiotics could increase the content of SCFA in intestinal tract (p<0.05,0.01).4. AG could increase the gene expression of HMG-CoA-R in liver of hamsters. And polydextrose, polyxylose, and inulin could increase the gene expression of LDL-R significantly (p<0.05).Comparative investigation on the effect of four species of prebiotics on the anti-oxidation possibility in hypercholesteremic hamsters was conducted through examining the indexes followed. It can be concluded that polydextrose, polyxylose, inulin, and arabinogalactan, could promote the activities of endogenous anti-oxidation enzymes and reduce the level of peroxidation in body, which attribute to the effect of preventing AS. Therefore, the four prebiotics could regulate the lipid of blood significantly.
