Objective: To investigate the expression and significance of Skp2、FAK in gastriccancer and their relationship; Analysis SKP-2,FAK and Hp(Helicobacter pylori) andHp-L(Helicobacter pylori L-form) infection in gastric cancer tissue. Methods:(1)usingreverse transcription polymerase chain reaction (RT-PCR) technique detection theexpression of Skp2, FAK mRNA in40cases of freshgastric carcinoma tissues andcorresponding normal gastric mucosa tissues (control group).(2)useingimmunohistochemical Elivision method detectionin protein expression of Skp2andFAK in120cases of gastric cancer tissues and40cases of the corresponding marginnormal tissues (control group).(3) Immunohistochemical Elivision method and Gramstaining method was used to detect the Hp and Hp-L in paraffin embedded tissueinfection.Results:(1)RT-PCR results show: Expression of Skp2,FAK mRNA in fresh gastriccarcinoma were significantly higher than the expression in normal tissue cutting edge inthe corresponding (P<0.05).(2)Immunohistochemistry showed:①Skp2protein positive expression rate in gastriccancer group (76.7%,92/120) higher than those in the control group (25%,10/40)(P<0.05), and the FAK protein positive expression rate in gastric cancer group (76.7%,92/120) higher than those in the control group (25%,10/40)(P<0.05).And Skp2, FAKprotein expression significantly increased with gastric cancer stage, depth of invasion,lymph node metastasis and distant metastasis(P<0.05), no correlation with patients sex,age, tumor size (P>0.05).(3) The results of detection of Hpand Hp-L: Type Hp-L in the group of gastric cancerthe immunohistochemical positive rate was79.2%(95/120), gastric cancer group therewere87Gram staining in detection of L, only13cases of simultaneous detection of Land Hp, detection rates were72.5%(87/120) and10.8%(13/120), the positive rate ofHp-L in gastric cancer group (i.e. Gram staining L positive and immunohistochemicalHp-L antigen expression and positive) was69.2%(83/120) higher than that of the control group37.5%(15/40)(P<0.05).(4)①The positive expression of Skp2, FAK protein ratio of Spearman rank correlationanalysis, results the positive expression of Skp2protein and FAK protein in gastriccancer related (r=0.362, P<0.05);②The HP-L gastric cancer group and the positiveexpression rates of Spearman rank correlation test rate and the positive rate of Skp2protein expression, FAK protein expression, results expression of HP-L was positivelycorrelated with positive and Skp2protein, FAK protein.Conclusion:(1) Expression of gastric cancer in the group of Skp2, FAK protein andmRNA were all up-regulated, suggest that they may be related to gastric carcinogenesis,development, and Skp2, FAK protein expression in gastric cancer group and the degreeof tumor differentiation, depth of invasion, lymph node metastasis and TNM stage, tosuggest that they are in the development of gastric cancer, and play an important role inthe process of invasion and metastasis.(2)Expression of Skp2and FAK protein werepositively related, suggesting that both play coordination role in gastric carcinogenesis,development.(3)Gastric HP-L infection and Skp2, FAK were positively correlated to,suggesting that the three may be involved in gastric carcinogenesis, development andmetastasis.
