| OBJECTIVETo explore the effects of Qili qiangxin capsule on myocardial fibrosisand expression of connective tissue growth factor(CTGF)in rats withchronic heart failure(CHF).METHODSCHF model was induced by partially banding abdominal aortic artery.Model rats were divided into model group(constant volume of normalsaline, CHF group),enalapril group(10mg/kg/d, ENA group),Qiliqiangxin capsule high-dose(1mg/kg/d, QLH group),medium-dose(0.5mg/kg/d, QLM group), low-dose groups(0.25mg/kg/d, QLL group)andsham-operation group(constant volume of normal saline,SHAM group).They were given medicine once a day for consecutive4weeks.Hemodynamic parameters and left ventricular weight index(LVMI)weredetected,Picric-Sirius Red(PSR)Polarimetry was used to observe collagenmorphology in left ventricular interstitial tissue,measure collagen volumefraction(CVF);the expression of CTGF in left ventricular tissue was evaluated by immunohistochemical staining and Western blot analysis.RESULTS(1)Compared with the sham-operated group,indexes of hemodynamicswere markedly anomalous (P<0.01) in CHF group rats. In rats treated byQili qiangxin capsule (1.0and0.5mg/kg/d) compared with those in CHFgroup, LVEDP and-dp/dtmax were significantly ameliorated (P <0.01or P<0.05).+dp/dtmax was significantly increased (P <0.01) in QLH groups.(2)Compared with the SHAM group, LVMI was significantlyincreased in CHF group rats (P <0.01); compared with the CHF group,LVMI were significantly reduced in QLH group,QLM group and QLLgroup (P <0.01).(3) Compared with the SHAM group, I CVF,III CVF and I/III CVFwere significantly increased in CHF group (P <0.01); compared with theCHF group, I CVF and III CVF were significantly lower in QLHgroup,QLM group and QLL group (P <0.01or P <0.05); I/III CVF wassignificantly lower in QLH group, QLM group (P <0.01).(4) Compared with the SHAM group, expression of CTGF protein wassignificantly increased in the CHF group (P <0.01); compared with theCHF group, expression of CTGF protein were significantly significantlydecreased in QLH group, QLM group and QLL group (P <0.01or P <0.05).CONCLUSIONSQiliqiangxin capsule attenuates ventricular hypertrophy in CHF rats, inhibits MF and improves cardiac function,which is dependent on its effecton expressions of CTGF in left ventricular tissue. |
