BackgroundIt is widely considered that white matter damage is the most important risk factor for cerebral palsy. Due to the impact of white matter damage, in-depth study for the repair mechanism is of great significance to reduce mortality and maim rate. Acupuncture as our motherland medical treasures, are very therapeutic for the cerebral palsy. EA is different from the traditional acupuncture, it combined the effects of acupuncture needle stimulation with physiological effect in order to cure the disease. This method not only improved the therapeutic effect but also expanded the scope of acupuncture treatment.ObjectiveThis project aims to observe and investigate the effects of cereb ral white matter nerve myelin sheath in the brain of the young offs pring of rats with intrauterine infection and the effects of galvano-ac upuncture. MethodsTo prepare the innate brain damage models:Injecting LPS (450μg/kg.d) to abdominal cavity of pregnant rats in17d and18d; injec ting equally sterile NS to abdominal cavity of contrasting pregnant r ats. the LPS young offspring of rats were randomly divided into the non-intervention groups and the galvano-acupuncture groups. The ga lvano-acupuncture groups start to train in d7. Using electron microsc opy to observe the cerebral white matter nerve myelin sheath thickn ess in d14、d28and stereological technique to measure and calculate the average thickness of cerebral matter nerve myelin sheath. Mean while.The treatment of EA:according to the references, we stimulate t he offspring rats of the EA group from the7th day with electro-acu puncture intervention. we stimulated the offspring rats with continuo us wave for10min once a day until rats grew to the28th day.We use transmission electron microscope to observe experiment result, five vision of each slice were taken randomly at20,000time s in magnification transmission electron microscope.Application stereological techniques to measure and calculate the average myelin thickness of white matter of offspringResults There were a large amount of neutrophilic granulocyte in the ut erus and placenta in LPS-treated group;1-day-old rat brain tissue pa thology test showed:white matter of LPS group, corpus callosum,int ernal capsule parts organizations osteoporosis, Glial cells gathered, C apsule of oligodendrocyte reduction, and can also be found vasodilat or, White matter capillary bleeding; there were no abnormal in NS group. The non-intervention groups and the galvano-acupuncture grou ps have a lower average thickness measurements than the control gr oups in cerebral matter nerve myelin sheath (P<0.01); the galvano-ac upuncture groups has a higher average thicConclusionsThe average thickness of myelin sheath is significantly decline d by the Intrauterine infection, which may be associated with oligod endrocyte injury and nerve fibers rupture. However, early electro-acu puncture intervention can effectively improve the average of the whit e matter myelin thickness.Our research proved that electro-acupuncture intervention are fav orable for the repair and regeneration of white matter myelin, which provided guidance significance for the further study of clinical ther apeutic value for children brain injury.
