Object ive: To investigate the photochemotherapeutic effect ofLED-activated MPPa on human giant cell tumor in vitro. To explore themechanism of photodynamic therapy. To provide the theoretical basis forclinical application.Methods: GCT-0404cells were treated with different concentrationsof MPPa (0ug/L、0.5ug/L、1ug/L、2ug/L、4ug/L) and were irradiated withdifferent light energy of LED (0J/cm2、3.2J/cm2、6.4J/cm2、12.8J/cm2).Then the OD values and survival rates of GCT-0404cells were measuredby MTT assay. After the treatment, the morphological changes ofGCT-0404cells were observed under electron microscope, and the celldeath was determined by Annexin V/PI double staining technique andanalyzed by flow cytometry. Observe the karyomorphism with DAPIstaining.Results: The giant cell tumor had a low survival rate after thephotodynamic therapy(p<0.05), when the dose of MPPa is2ug/L and thelight energy of LED is12.8J/cm2. Electron microscope found that the cellswere shrunk, the cytoplasm were colored deeply, the nucleus chromatin were condensation and the The cell membrane is completely. The DAPIstaining found the heteropyknosis of nucleus. The flow cytometry showedthat advanced stage apoptosis cells and necrosis cells.Conclusions: It had lethal effect of LED-activated MPPa on humangiant cell tumor in vitro. The mechanism of it is apoptosis and necrosis. Itprovides the reference for next research.
