Objective: To clarify the effects of curcumin (CCM) combined withHydroxycamptothecin (HPCT) on proliferation and apoptosis of humanbladder cancer cell line BIU-87and discuss its mechanism initially.Methods: Expression of YAP was detected by IHC in bladder cancerand para-cancer; the human bladder cancer cell BIU-87was cultured; theproliferation and apoptosis rates were performed by CCK-8assay and flowcytometry (FCM) after the cells were treatment by CCM or HPCT andcombined with the two drugs; the expressions of YAP, TEAD1mRNA andprotein were detected using RT-PCR and western blot, respectively.Results: The proliferations of the BIU-87cells were inhibited by CCMor HPCT effectively and had correlation with concentration and time; Theinhibitory rate of combination treatment with CCM or HPCT wassignificantly higher than that of CCM or HPCT alone (P<0.05); It wasconfirmed that apotosis rate of cells treatment by CCM combined withHPCT shows significant increasing than that of CCM or HPCT alone byflow cytometry (P<0.05); expression of YAP, TEAD1mRNA and proteinwere inhibited by treatment with CCM or HPCT, while inhibitory rate increased more by treatment with the two drugs through RT-PCR andwestern blot; there were significant differences between treatment groupsand control (P<0.05) and combined group as well as treatment groups alone(P<0.05).Conclusion: CCM combined with HPCT had obvious synergisticeffects on inhibiting the proliferation of BIU-87cells and enhancing theapoptosis. The mechanism of effects on inhibiting the proliferation may bethat expression of YAP in Hippo signal pathway was inhibited by CCMcombined with HPCT, and even interfered the expression of TEAD1, soapotosis occurred.
