Objective: Ketamine is still widely used in clinical anesthesia work, isnow only in the use of ring c has piperidine drugs.In recent years there aremany experts and scholars study found that ketamine in addition to alreadyknown as the sedation and analgesia anesthetic effect outside, still has asignificant anti-inflammatory effect.Other scholars in the inflammationresearch found that nuclear factor-κB in inflammation plays a key role.Sothe effect of ketamine in anti-inflammatory and the inhibition of nuclearfactor-κB whether there is a certain relationship?The design of this study isto investigate the effect of ketamine on lipopolysaccharide-inducedexpression of the translocation of nuclear-kappaB(NF-κB)in nuclei ofhuman umbilical vein endothelial cell and explore it’s anti-inflammatoryaction.Methods: Human umbilical vein endothelial cells(HUVECs) inDMEM medium, training, transfer of culture, vaccination to petri dishes,until the attached growth to70~80%fusion rate was used to experiment.The human umbilical vein endothelial cellswere divided into4groups.(1)C group for blank control group,in which isometric DMEMculture medium was added to the DMEM culture medium.(2)L group inwhich LPS1μg/ml was added to the DMEM culture medium.(3)Ketaminegroups were divided into2subgroups according to the concentration ofketamine added:3μg/ml+1μg/ml LPS(KL1)、30μg/ml+1μg/mlLPS(KL2).We detected the tanslocation and expression of NF-κB p65inendothelial cell nuclei with the method of Western blotting、RT-PCR andimage analysis system of HPIAS2000.Results: The tanslocation and expression of NF-κB p65in endothelialcell nuclei induced by Lps were reduced in group KL1and KL2(P<0.05).Conclusion: The tanslocation and expression of NF-κB p65inendothelial cell nuclei can be induced by Lps,and ketamine can suppress theactivation.There was positive correlation of concentration between groupKL1and KL2. The anti-inflammatory action of ketamine may act by thetranslocation of NF-kappaB.
