Objective:To copy the rat model of airway mucus hypersecretion to acrolein method, by observing the particle heat phlegm medicine Xingpa Zhike of inflammatory cells, goblet cells stained positive rate and serum neutrophil elastase (NE) and pathological changes in lung tissue and intervention, in order to investigate the mechanism of airway mucus hypersecretion of Xingpa Zhike granules control rats.Methods:48healthy male SD rats, were randomly divided into6groups, with8rats in each group, respectively, the blank group (group A), model group (group B), low dose Xingpa Zhike granule group (group C), middle dose Xingpa Zhike granule group (group D), high dose Xingpa Zhike granule group (group E) and ambroxol group (group F). Group B, C, D, E, F with3.0ug/ml acrolein atomization inhalation, group A normal saline,6h/d,5days weekly, the end of the twelfth day.Group C, D, E, F in acrolein atomization1day began with4.4g crude drug/kg,8.8g crude drug/kg,17.5g crude drug/kg,40mg ambroxol tablet/kg dissolved in3ml normal saline by gavage, group A and group B were given normal saline, one times every day, duration12days,12days later, rats in the model group and the middle lobe of the right lung pathological section examination revealed a bronchial lumen and wall infiltration of inflammatory cells, and hyperemia, edema, inflammatory exudate obstruction, bronchial epithelial and cilia shedding, goblet cell hyperplasia hypertrophy, alveolar structure disorder disappear, alveolar wall thinning or fracture, massive fibrosis real variable, lung volume reduction cystic dilatation, alveolar cavity enlargement, some fused into pulmonary bulla judged model was successfully established, after the success of modeling.Bronchoalveolar lavage fluid (BALF) for cell sorting and counting, right lung lobe HE staining for understanding pathological changes, AB-PAS in lung tissue staining goblet cells stained positive rate detection of serum NE, intraluminal mucus secretion and ELISA method, etc.Results:(1)model replication:through pathology airway mucus hypersecretion in rat model was duplicated successfully.(2)the BALF cell sorting and counting middle and high doses of xingpa Zhike granules group total cells, eosinophils, neutrophils, lymphocytes decreased as compared with the model group, in the statistical, the difference was significant (P<0.05orP<0.01);â‘¡AB-PAS staining goblet cells stained positive rate of low dose Xingpa Zhike granules group, middle dose Xingpa Zhike granules group, high dose Xingpa Zhike granules group, ambroxol group than in model group of goblet cells stained positive rate decreased, the differences were statistically significant(P<0.01); compared with the model group decreased dose Xingpa Zhike granules group, high dose Xingpa Zhike granules group, ambroxol group serum NE content of serum in NE obviously, there is significant difference (P<0.01); the HE staining bronchial epithelial mucosa of rats in model group under the gland hypertrophy, hyperplasia, inflammatory cell infiltration and secretion blocking, cilia arranged in disorder; low dose Xingpa Zhike granules group than in model group had no obvious change; reduce dose Xingpa Zhike granules group, seen in the trachea bronchial inflammation cell infiltration; high dose Xingpa Zhike granules group, ambroxol group of bronchial wall thickening was no, no stenosis, no discharge clogging, even visible Shao Xuyan Cells.Conclusion:(1)close airway inflammatory cells, goblet cell hyperplasia and blood NE and chronic airway inflammatory disease of airway mucus hypersecretion relationship.(2)Xingpa Zhike granules by airway mucus hypersecretion clearing heat and resolving phlegm method in rats, its mechanism of action and reduce airway inflammatory cells, goblet cells stained positive rate, serum NE level.
