Objective:Through the research on creatine-kinase(CK), creatine-kinase-MB(CK-MB),lactate dehydrogenase(LDH), aspartate-amino transferase (AST) and α-HBDH;endothelin(ET),6-Keto-PGF1α, endothelial nitric oxide synthase(eNOS), nitric oxide(NO)and thromboxane(TXB2) in plasma as well as malondialdehyde (MDA) and superoxidedismutase (SOD) in myocardial tissue of rat models of acute myocardial infarction treatedwith Gualoupi injecta, to evaluate the effect of Gualoupi injecta in myocardial infarctionprotection and provide experimental evidence for the clinical application of trichosanthespericarpium.Methods:To establish the animal model of acute myocardial infarction by ligating the leftanterior descending coronary branch. To get the blood and myocardial tissue after beinganesthetized, the myocardial tissue was dyed with triphenyltetrazolium chloride (TTC).The degree of myocardial infarction was evaluated by the infarction area and the infarctionrate. To evaluate the myocardial injury, levels of CK, CK-MB, LDH, AST and α-HBDH inthese different groups of rats were detected by semi-automatic biochemical analyzer; thespectrophotometer meter was used to measure the content of MDA and SOD in myocardial tissue, to find out the mechanism of trichosanthes pericarpium in protecting myocardialinfarction, and measure the content of ET, eNOS, NO,6-Keto-PGF1α and TXB2withELISA.Results:Compared with that in the model group, the myocardial infarction rate of rats inthe Trichosanthes group decreased evidently, the activity of CK, CK-MB, LDH, AST andα-HBDH were also obviously lower (P<0.05or P<0.01). The activity of MDA wasreduced, and that of SOD in the myocardia increased observably. The content of NO、eNOS,6-ketone-PGF1α increased and that of ET、TXB2decreased.Conclusion: Gualoupi injecta could protect myocardial infarction in rats effectively, andits mechanism may be related to reducing the release of cardiac enzymes, inhibiting thelipid peroxidation of ischemic myocardia, improving the ability of resisting oxygen freeradical damage, promoting vasodilation, inhibiting vasoconstriction, inhibiting plateletactivation and aggregation as well as reducing thrombosis and so on.
