Objective: By adopting the modern molecular biology technology, We discuss therelationship between ACE2and Nrf2and diabetic myocardium injury,and the possiblemechanism.Method: Adopt STZ to induce diabetic rats,observe the situation of the diabeticrats;collect the heart of the rat、fix、cut into slices、HE stain and Masson stain,observe theshapes under light microscope,observe the myocardial cell and interstitial tissue changes ofmyocardium,and account the CVF through image pro-plus6.0image handlingsoftware,using PCR technology assay the expression of ACE2and Nrf2inmyocardium,guess the possible mechanism between them,collect patients from outpatientsand inpatients,40patients diabetic cardiomyopathy complete the trial,grouping them inrandom into trial group and control group,there are no difference between the two groupsin age、 gender and the pathogenesis, the trial group was given Membranaceusdecoction,while the other group did not,the two groups accept basic treatment for diabetiecand heart failure,measure the blood glucose、the level of the heart function and constrictionof left ventricular through echocardiogram,assay the multi-effect and Chinese medicalsymptoms,guess the clinical interference of Membranaceus.Results: Compared with the normal rats,the injury of myocardium and the collagen ininterstitial tissue of myocardium increased in diabetic rats,compared with diabetic rats, theinjury of myocardium and the collagen in interstitial tissue of myocardium reduced indiabetic rats feeded with Membranaceus, compared with the diabetic rats withoutMembranaceus,the expression of ACE2and Nrf2reduced in the heart;during the clinicaltrials,patients given Membranaceus decoction have improved activity tolerance. Conclusion: The expression of ACE2and Nrf2reduced in the heart in diabeticrats;the expression of ACE2and Nrf2may involved in the protection in diabebticmyocardium;ACE2may down-regulate the expression of Nrf2through Ang Ⅱ-AT1R-NADPH oxidase pathway;Membranaceus can interfere the injury of diabeticmyocardium with little blood glucose down regulation, Membranaceus decoctionshave.some effect on improving the activity tolerance of patients with diabeticcardiomyopathy.
