Objective: By observing the influence of matrix Gla protein on human vascularsmooth muscle cell and human MG-63cell intervened by four anti-osteoporosisagents(vitamin K2, PTH,1,25(0H)2D3, Alendronate), we try to find the possible roleof MGP on pathogenesis of osteoporosis and possible role of osteoporosis treatmentdrugs on prevention of arterial calcificationin.Methods:1.Choose human vascular smooth muscle cell and classic osteoblastphenotypic characteristics of osteoblasts model into osteosarcoma MG-63cell tosubculture.2. All cells were treated with different concentrations of vitamin K2, PTH,1,25(0H)2D3, Alendronate by intervention cultured24h: Vitamin K2(10-7,10-6,10-5mol/L),1,25(0H)2D3(10-10,10-9,10-8mol/L), PTH (10-9,10-8,10-7mol/L),Alendronate (10-6,10-5,10-4mol/L). Set up a blank check for all medicine.3.Extractthe total protein on vascular smooth muscle cell and MG-63cell after24h, then detectthe change of the expression of MGP by western-blot.Results:1. The influence of the expression of MGP in vascular smooth musclecell by treadted by Vitamin K2, PTH,1,25(0H)2D3and Alendronate: Vitamin K2,PTH,1,25(0H)2D3and the Alendronate all can stimulate the expression of MGP inhuman vascular smooth muscle cell, and as the graded increasing of drugconcentration,the expression of MGP is significantly higher.2. The influence of theexpression of MGP in MG-63cell by treadted by Vitamin K2, PTH,1,25(0H)2D3and Alendronate:Vitamin K2, PTH,1,25(0H)2D3and the Alendronate all canstimulate the expression of MGP in MG-63cell, and as the graded increasing of drugconcentration, the expression of MGP is significantly higher.Conclusion: It can be confirmed that anti-osteoporosis agents can affect theexpression of MGP in human vascular smooth muscle cell and the expression of MGPin MG-63cell by western-blot technique. Anti-osteoporosis agents like vitamin K2,PTH and active1,25(0H)2D3, alendronate may promote the expression of MGP invascular smooth muscle cell and MG-63cell in a dose-dependent manner. We thinkthat anti-osteoporosis agents may become a new research direction for the prevention and treatment on arteriosteogenesis. MGP always may be Involved in thepathogenesis of osteoporosis,and it may be a target for the treatment of osteoporosis.
