Objectives:Observation Tanreqing injection (TRQ) on endotoxin (Lipopolysccharide,LPS)in mice with Acute lung injury,Acute lung injury,ALI),arterial blood gas analysis,lung index, serum NO content and Caspase-3(Cysteinyl aspartatespecificproteinases-3, Caspase-3) protein expression changes,etc,the influence of the TRQALI protective effect and its mechanism are discussed.Methods:Health clean ICR mouse30(female and male do not arrest,5weeks of age,bodyweight20~25g), according to divides into following three groups completelystochastically (each group of10);1. Physiological saline (Normal saline, NS) group.2. LPS induced acute lung injury (ALI group),3. LPS+TRQ group (TRQ group).TheALI group and the TRQ group the drop note (LPS4mg/kg,2ul/g) prepare the acutelung damage model by the trachea in, the TRQ group the drop pour LPS while thetrachea, immediately gives the TRQ inoculation fluid4ml/kg tail intravenousinjection.After3groups choose the technique6h are the observation points, observeseach group of lung index, the blood serum NO content and the comparison arterycourage vigor analysis result; Observation lung organization pathology morphologychange; Immunity histochemistry examination lung Caspase-3protein expressionsituation.Results:1. General performance: Drip drugs after6h, NS group in the process of theexperiment did not see abnormalities; curled up, ALI group mice chills to the cold,speeding up breathing, respiratory distress began to emerge, and even appear purplepurple; TRQ group to speed up, breathing frequency range, no purple purple;2. Lung index results: ALI lung index higher than that of NS group (P <0.05);Compared with ALI group, the TRQ group decreased lung index (P <0.05); TRQ group compared with NS group, lung index, both no statistical difference (P>0.05).3.Artery courage vigor analysis determination: NS group, the ALI group and thePH of the TRQ group, is no statistical difference (P>0.05); ALI group PaO2declinedsignificantly in the NS group (P <0.01); Compared with ALI group, PaO2TRQgroup increased significantly (P <0.05); TRQ group compared with NS group ofPaO2, both no statistical difference (P>0.05).4. In blood serum NO content determination: Compared with NS group, ALIgroup and increased serum NO content of the TRQ group (P <0.01); Compared withALI group, the TRQ group serum NO levels decreased significantly (P <0.01).5. Lung organization pathology morphology change: Then NS group of alveolarducts and alveolar walls structure integrity, alveolar lumen is bright and clean,alveolar walls and no inflammatory cell infiltration in the stroma; ALI grouppathological changes obviously, wide alveolar septa, alveolar lumen narrowed,inflammatory cells exudation, mainly neutral cells and lymphocytes; TRQ grouppathological changes is lighter, lung interval widened slightly, slightly narrow alveoli,a small number of alveoli and the blood vessel walls surrounded by lymphocyteinfiltration, damage degree of the ALI group obviously reduce.6. The lung organizes the Caspase-3expression change: NS group of caspase-3expression in lung tissue are rare. ALI group of caspase-3expression in lung tissues ishigher than other groups, for the strong positive. TRQ group of caspase-3expressionin lung tissue was higher than NS group, below the ALI group, weakly positive.Compared with NS group, ALI group and TRQ Caspase-3expression positive rateincreased (P <0.01); Compared with ALI group, TRQ group of Caspase-3expressionpositive rate decreased significantly (P <0.01).7. Lung index and between blood serum NO content relevant analysis: Therewas a positive correlation between the changes of lung index and the content of serumNO (r=0.516, P=0.003, P<0.001).8. Artery blood oxygen partial pressure and between blood serum NO contentrelevant analysis: There is a negative correlation between changes in arterial oxygenpartial pressure and the content of serum NO (r=-0.545, P=0.002, P<0.01). Conclusions:1.NO in the pathogenesis of ALI induced by LPS plays an important role in theprocess.2.TRQ on LPS-induced ALI has a protective effect, probably by reducing serumNO content, reduced the expression of Caspase-3protein, thereby reducing the lungtissue damage.
