The Effective Composition Analysis And Efficacy Evaluation Of Longchai Decoction

Composite formula of Chinese Medicine (CFCM) contains a large number of chemical compositions which are the material basis for the clinical efficacy. For further mining the compatibility and mechanisms of action, the efficacy of CFCM is supposed to be clarified. So the research on effective substance basis is momentous for the modern study of CFCM.This work was supported by "science and technology foundation construction plan" of Jiangsu province (No. BM2009903), Longchai Decoction used for the treatment of chronic hepatitis B comes from the commonly used decoctions of Jiangsu Province Hospital of Traditional Chinese Medicine. It is, mainly composed of eight kinds of chinese herbal medicines, such as Bupleuri Radix, Scutellariae Radix, Glycyrrhizae Radix et Rhizoma, etc. Against the pathogenesis of hepatitis B, the Longchai Decoction can make human body recover when it is capable of improving the clinical symptoms of damp-heat and toxin by heat-clearing and detoxicating, regulating spleen and stomach.The main task is to study the chemical constituents of Longchai Decoction, It preliminary analyse the changes of the chemical composition before and after compatibility, which confirmed that the differences in the chemical compositions between traditional decoction and mixed dividual decoctions were very obvious. The content of the index components was increased to some extent in traditional decoction. The analysis of chemical compositions in compatibility supplied the basis for elucidating the scientific connotation of traditional decoction theory for CFCM. By animal experiments, preliminary analysis on the mechanism of the curative effect of chronic hepatitis B was obtained. The research mainly includes the following aspects:1. Literature study:Literatures related to Longchai Decoction up to now are studied thoroughly.2. Quality evaluation of single Chinese herb:Quality evaluation of each single Chinese herb is based on the analysis of Longchai Decoction, only to ensure their stable source, the analysis of Longchai Decoction is meaningful. The content analysis of active ingredients about each single Chinese herb is carried out by HPLC or HPCE. The results demonstrate that Longchai Decoction is composed of black nightshade from lianyungang, Sedi Herba from hubei, Herba Hedyotidis Diffusae from guangdong, etc. It is regarded as the object of the effective component analysis and efficacy evaluation.3. Effective component analysis of Longchai Decoction:According to the results of the second part of the experiment, simultaneous determination of the contents of a variety of active ingredients is carried by HPLC, HPCE, UPLC-Q-TOF-MS and HPLC-T-Q-MS. To probe into the effectiveness and safety of Longchai Decoction, experiment using ICP/MS is designed to research water soluble inorganic elements of Longchai Decoction, which is considered the clinical use characteristic. The results show that the contents of active ingredients were richer in traditional decoction than that in mixed dividual decoctions, which may be caused by the solubilization of varieties of medicinal ingredients in the decocting process. These changes may also be the material basis of the efficacy of Longchai Decoction.4. To establish the fingerprint chromatogram of Longchai Decoction:Fingerprint is based on the study of chemical constituents of Traditional Chinese Medicine, used for evaluation of its authenticity and stability. Fingerprint can also comprehensively reflect the type and amount of chemical components, and then conduct an overall description and evaluation of the quality of TCM. The fingerprint chromatogram of Longchai Decoction is discussed in this part of the experiment by HPLC, providing quality control means in order to ensure that the inherent quality of Longchai Decoction. The HPLC fingerprints of Longchai Decoction were established for evaluating the effective substance basis with different commodities. Its main chromatographic peaks of the HPLC fingerprints of Longchai Decoction were identified in this experiment, it can be used for the quality control of Longchai Decoction.5. Efficacy evaluation of Longchai Decoction:Through the study on the inhibitory effect of hepatitis B virus and the promoting effect of hepatocyte growth, to explain the mechanism of hepatitis B on Longchai Decoction. The results show that the Longchai Decoction has a certain role to protect the liver.
