Aims:Metabonomics is a new field on-omics in post-genome era. Stimuli including drugs, foods, environment and etc. can lead the organisms to adjust, eventually changing the whole metabolome, which is the end of all kinds of biological process. By investigation on the metabolites in the serum of ascites tumor bearing rats after treatment with Xiao’ai Jiedu Prescription, we aim to know the therapeutic effects and mechanisms of ascites due to hepatic cancer.Methods:1) We use Wistar rats subcutaneous injected with Walker-256cell line to model ascites induced by hepatic cancer. We then treated these rats with Xiao’ai Jiedu Prescription and normal saline administration by gavage, isolated the serum ten days later from the whole blood.2) Based on GC/TOF-MS platform, we established the metabolic fingerprint of those serum samples.3) We last statistically analyzed the content of metabolites in the serum of those two groups by t-test (significant, P<0.05; obvious significant P<0.01).Results:The changes in the metabolic fingerprint between the model, treatment and control groups are obvious. After database retrieval, we can deduce that the levels of2,3-Dihydroxybutanoic acid and2,4-Dihydroxybutanoic acid in the model group increase, and the levels of3-Hydroxybutyric acids beta-Alanine、Aspartic acid、Glucose、Cystine in the treatment group also increase. Therefore, we conclude that the Xiao’ai Jiedu Prescription can regulate the metabolism of amino acid and carbohydrate.Conclusions:Through the comparison of the serum metabolites in each group, we conclude that the Xiaoai Prescription can treat the ascites induced by malignant tumors. From the research, we can analyze the metabolites differences from patients with tumors after treatment with Chinese medicine, find the putative targets, in order to provide the evidence for diagnosis and clinical treatment.
