Objective:To observe the effects of metformin on proliferation and apoptosis of hepatoma cell line Huh-7, and to explore the mechanisms.Methods:(1)Culture hepatoma cell line Huh-7in different concentrations of insulin and glucose in vitro, and treat with different concentrations of metformin(0,5,10,20mM), then detect the proliferation rate with MTT assay.(2)Treat hepatoma cell line Huh-7with different concentrations of metformin(0,10,20mM), and then detect early apoptosis rate with flow cytometry.(3)Treat hepatoma cell line Huh-7with different concentrations of metformin(0,10,20mM), and then detect the expression levels of STAT3and p-STAT3.Results:(1) Metformin inhibits the proliferation of hepatoma cell line Huh-7that are cultured in different concentrations of insulin and glucose in vitro in a dose-dependent manner(p<0.05).(2)Metformin induces the apoptosis of hepatoma cell line Huh-7in a dose-dependent manner(p<0.05).(3) Metformin doesn’t affect the expression of STAT3in hepatoma cell line Huh-7, but reduces the phosphorylation of STAT3(p<0.05).Conclusions:(1)Metformin inhibits the proliferation of hepatoma cell line Huh-7in vitro in different concentrations of insulin and glucose, and the effect is in a dose dependent manner.(2)Metformin induces the apoptosis of hepatoma cell line Huh-7in a dose dependent manner.(3)Metformin inhibits the phosphorylation of STAT3in hepatoma cell line Huh-7, and it may induce apoptosis at the transcriptional level.
