| Objective:Purpose To explore the relationship of three single nucleotide polymorphisms(SNP) loci in FOXP3gene (rs2232365, rs3761547, rs3761548) and diabete mellitus after kidney transplantation or posttransplantation diabete mellitus(PTDM) by experiment and clinical analysis, and to investigate whether the genetic polymorphisms of FOXP3contributes to the PTDM. Then to prevent the PTDM in clinical practice.Methods:167patients who spend the whole post-operation follow-up period in the organ transplantation out-patient clinic in our hospital were divided into PTDM and non-PTDM groups. All the patients’DNA samples were collected, and PCR-RFPL was applied to detect3SNP loci rs2232365, rs3761547and rs3761548; statistic method was taken to analyse the results of the experiment to investigate the relationship of genetic polymorphisms in FOXP3gene and new-onset diabete mellitus after kidney transplantation.Results:1,8patients in PTDM group and the other159patients in non-PTDM groups, and the incidence of new-onset diabete mellitus after kidney transplantation was4.79%in our transplantation centre.2, The allele frequence distribution of loci rs2232365in gene FOXP3were A64.97%, G35.03%. Genotypic frequence were AA58.68%ã€AG12.57%ã€GG28.75%; the allele frequence distribution of loci rs3761547in gene FOXP3were A79.04%, G20.96%. Genotypic frequence were AA75.44%, AG7.19%, GG17.37%; the allele frequence distribution of loci rs3761548in gene FOXP3were A14.37%, C85.63%. Genotypic frequence were AA10.77%ã€AC7.19%ã€CC82.04%. 3, According to chi square test of the allele frequence and genotypic frequence, the P values of the three SNP loci in the FOXP3gene were0.082,0.670,0.245, all>0.05, which were in coincidence of Hardy-Weinberg law(law of genetic equilibrium), and the group representativeness confirmed.4, The difference of demography statistics of the two groups, including age, sexuality, DM family history, dialysis time, dialysis mode, BMI, whether HCV infection, choice of FK506, blood fasting sugar before operation, has no statistic significance (P>0.05)5, The difference of genotypic frequence distribution of the two loci rs2232365and rs3761548in gene FOXP3had no significant difference, while the difference of genotype AA and genotype AC of rs3761547had statistic significance(P=0.033, P=0.007). The frequence of genotype AG was higher in the PTDM than that of non-PTDM group(OR=10,95%CI=2.057-48.624, P=0.004).Conclusions:1, The incidence of new-onset diabetes mellitus after kidney transplantation was4.97%in our oran transplantation centre, which is a low incidence.2, The SNP of loci rs2232365,loci rs3761548in gene FOXP3has no significant relation with PTDM.3, The loci rs3761547in gene FOXP3probably has relation with the incidence of new-onset diabetes mellitus after kidney transplantation, and the genotype AG may become the predisposing genes of new-onset diabetes mellitus after kidney transplantation. |
