Objective To study the effect of silibinin on lung metastasis of ACC-M, and to investigate the inhibitory mechanism.Method20nude mice which were injected0.2ml RPMI1640with ACC-M cells (106/ml) via caudal vein, were divided at random into two groups:experimental group and control group.The mice in experimental group were given0.8ml silibinin solution (lmg/ml) diluted by RPMI1640by intraperitoneal injection after24hours, while the ones in control group were given only0.8ml RPMI1640. Six weeks later, mice were euthanatized. The weight of lung and the number of lung metastasis focals was measured respectively. Immunohistochemistry, reverse transcription polymerase chain reaction (RT-PCR) and immunoblotting were used to detect the expression of LC3in the lung metastasis focals of adenoid cystic carcinoma.Results The lung’s weight of the mice in control group was (4.20±0.6) g, while the one in experimental group was (4.08±0.11) g. The number of metastasis focus on the left lung and the right lung of the mice in control group was (9.29±5.91) and (10.71±7.83) respectively, while the one in experimental group was (3.11±3.76) and (2.22±3.46) respectively. Compared with the control group, the lung’s weight and the number of metastasis focus on both left and right lung of mice in experimental group were all significantly less (P<0.05). The expression of mRNA of LC3in experimental group was (0.784±0.103), which was significantly higher than control group (0.685±0.077),(P<0.05). The expression of protein of LC3-Ⅰ and LC3-Ⅱ in experimental group was (0.446±0.093;0.567±0.070), which was significantly higher than control group (0.276±0.096;0.418±0.098),(P<0.05). The result of immunohisto-chemistry reavealed the expression of LC3in experimental group (80%) was significantly higher than control group (30%),(P<0.05).Conclusions Silibinin can inhibit lung metastasis of ACC-M in nude mice. The mechanism may be that silibinin can induce autophagy of tumor cells.
