| ObjectiveTo observe the effects of novaferon on the expression of TNF-a and IL-13in murine intestinal mucosa induced by DSS with ulcerative colitis and its possible mechanism. In order to provide experiment basis for biological treatment and effective method for clinical treatment of ulcerative colitis.Methods:70specific pathogen free, about8week-old female BALB/c mice (weight20.0±2.0g), were randomly divided into7groups:normal, model, mesalazine control, prednisone control, novaferon low-dose,novaferon middle-dose, novaferon high-dose group.10mice were involved in each group. The normal group-mice were fed with distilled water. Intervention group-mice were fed with4%dextran sulfate sodium (DSS, MW:36000-50000) freely for7continuous days to complete ulcerative colitis model. At the same time, every experimental group was orally administrated physiological-saline,physiological saline, mesalazine (40mg/ml), prednisone(5mg/kg), novaferon0.1ug/mlã€0.2ug/mlã€0.3ug/ml intraperitoneal injection respectively.During the experiment, general situation, daily weight, stool trait and occult blood were recorded,and mice were killed on the8th day. Isolated the whole colon, and taken piece of colon formaldehyde fixed, paraffin embedded, Hematoxylin and Eosin (H&E) staining.The following indices were used to assess the effect of ulcerative colitis model:disease activity index(DAI),colon length, histological scores. Immunohistochemistry (IHC) was used to measure the expression of TNF-a and IL-13in colonic mucosa.Results:1.BALB/c mice treated with DSS solution developed diarrhea, mucous stool and bloody stool.With days,DAI scores of all Intervention group were increasing.novaferon can improve the general situation of the mice.Compared with model group, the DAI scores, colon length, histological scores were statistically significant(P<0.05).2.In mesalazine control and novaferon high-dose group,the expressions of TNF-aand IL-13were both decreased, compared with model group(P<0.05).Conclusion:The manifestation s of DSS-induced acute colitis mice were similar to clinical symptoms of UC patients.Novaferon had therapeutic effects on the acute UC mice effectively. Its possible mechanism may be related to the the decrease of TNF-a and IL-13. |
