BackgroundLung cancer is the leading cause of cancer-related mortality throughout the world.Because of the insidious onset of lung cancer,80%of patients diagnoses in the advancedstage. Although efficacy of treatments can improve survival of some patients, the overallsurvival is still depressed. Therefore, explore new lung cancer treatment become a pressingproblem. Autophagy is a eukaryotic cell specific phenomenon. Beclin-1as importantregulatory factor of autophagy play important roles in the tumorigenesis and developmentof cancer. Beclin-1is a tumor suppressor gene in lung cancer, the down regulation ofBeclin-1cause autophagy and apoptosis defects which contribute to the occurrence anddevelopment of lung cancer. However the relationship of Beclin-1and lung cancerhistological type, degree of differentiation, lymph node metastasis, distant metastasis,gender, age, chemotherapy and other clinical parameters are rarely reported, it is importantto explore the relationship of Beclin-1expression and clinical parameters in non-small celllung cancer.ObjectiveTo investigate the expression of an important autophagy related factor beclin-1innon-small cell lung cancer (NSCLC) and its clinic significance. Explore the relationship ofBeclin-1expression and clinical factors or chemotherapy efficacy analysis.MethodsOver150cases of non-small cell lung cancer patient’s cancer tissues and adjacenttissues was made in tissue chip. The expression of Beclin-1is obtained byimmunohistochemical method in lung cancer and adjacent tissues. Cases to analyze therelationship between Beclin-1expression and lung cancer in different clinical parameters or survival analysis were performed. Select52clinical stage III-IV patients with localresection surgery and definite pathological diagnosis, cisplatin-based treatment. Lung cancertissue, chemotherapy and chemotherapy evaluation, survival time, cases of the relationshipbetween the different clinical parameters of Beclin-1expression and other relevantinformation with non-small cell lung cancer and survival analysis.ResultsThe positive expression rate of beclin-1in cancer tissues (26.00%) was significantlower compared to the adjacent tissues (82.00%)(P <0.05). It was associated withpathological types of lung cancer. In squamous cell carcinoma, the positive expression rateof beclin-1was (34.67%), which was significant higher than that of (17.33%) inadenocarcinoma. However, no evidence showed correlations between expression level rateof beclin-1and gender, age, degrees of differentiation, lymph node metastasis and distantmetastasis(P>0.05). Line kaplanMeier survival analysis, log-rank non-parametric testshowed that the patients with negative expression of beclin-1in cancer tissues getsignificant lower survive rate compared to the patients with positive expression (P<0.05).52cases of III-IV of lung adenocarcinoma patients’ overall response rate is46.15%,the Beclin-1positive expression in30cases (57.70%), negative expression in22cases(42.30%). Beclin-1expression and chemotherapy response has a statistically significantdifference (χ~2=13.099,P=0.043). Line survival analysis of Beclin-1expression intensityand patients survival period was significantly related (χ~2=9.968, P=0.007), but the patient’sgender (χ~2=0.266, P=0.606), age (χ~2=0.009, P=0.924), Clinical stage(χ~2=1.049, P=0.306),pathological type(χ~2=0.791, P=0.374)and survival in patients with no significantcorrelation.ConclusionThe positive Beclin-1expression rate in lung cancer tissues was significantly lowerthan that in the adjacent tissues, the positive expression rate of lung squamous cellcarcinoma was significantly higher than the positive expression rate of adenocarcinomalung cancer; survival analysis found survival with Beclin-1negative expression innon-small cell lung cancer was significantly lower than that with positive expression. It point out Beclin-1may be auxiliary indicator of non-small cell lung cancer pathology typejudgment.Beclin-1expression and chemotherapy response in patients with III-IV stage lungadenocarcinoma is a statistically significant difference, Beclin-1expression of patient wasno significantly related to gender, age, histological type,but was significantly related tooverall survival. Beclin-1may be used as the efficacy of chemotherapy predicted orevaluation of prognosis.
