| The realgar comes from the mineral sulfide realgar family, the main contain is arsenic disulfide (As2S2or As4S4). It was first recorded in the "Shen Nong Ben Cao Jing", and it has two thousand years history of clinical application in China. Realgar is one of the toxic drugs,its frequently lead to toxic reactions because of the inappropriate use,that make the status of seriously ill under light dose and drug isn’t symptomatic during its clinical application. This article focuses on study the content of As2S2and As2O3in realgar samples by different processing methods, and the spectral characteristics of different realgar samples. Under the premise of clarify the chemical composition of each sample and its spectral characteristics, we carry out some samples’acute toxicity experiments and anti-tumor research. Expect to provide experimental support for realgar’s clinical use.1.The research on contents variation of As2S2and As2O3in different realgar processed products(1) Processing three batches of realgar followed by water, acid water and alkali water, determine the rate of processed products and the weight of residue. The rate of recover are water processed> acid processed> alkali processed, residue weight are on the contrary.(2) We determine the contents of As2S2and As2O3in different realgar samples refer to "Chinese Pharmacopoeia"2010edition which under the realgar’s content determination and Appendix IX F second method. the content of As2S2(Guo ji shang mao cheng and Xin he hua) are alkali processed> water processed> acid processed>original; the content of As2S2(Hu nan yao fang) are acid processed> alkali processed> water processed>original:the content of As2O3are acid processed <alkali processed<water processed<original.Considering the rate of the processed products, weight of the residue, the content of As2S2and As2O3comprehensive. we"d better to choose water processed or acid processed, and abandon alkali processed.2. The research on spectral characteristics of realgar(1) Determined three batches of original realgar and processed products which corresponding to Xinhehua and Hunan yao fang(a total of ten samples) by X-ray diffraction, then obtained the corresponding X-ray diffraction spectra and diffraction peak data, and informed crystal structure of the sample described above. The results showed that there had differences in crystal structure between Hunan yao fang and other two groups of realgar; the crystal structure of original realgar was the same as its processed products, illustrated that the crystal structure of realgar won’t be changed by routine laboratory processing methods.(2) Identification three batches of original realgar and a number of realgar pieces by Fourier transform infrared spectroscopy, obtained the IR spectra. The results showed that there was no characteristic absorption peak at range of4000cm-1~400cm-1wave number in realgar infrared spectrum. This infrared that infrared spectroscopy isn’t applicable to identify the characteristics or quality control in realgar.(3) Identification three batches of original realgar and a number of realgar pieces by Laser Raman spectroscopy. There appeared obvious absorption peak at the wave number location of182cm-1,221cm-1,234cm-1,271cm-1,342cm-1in all four samples, this had also been confirmed on the second derivative Raman spectroscopy. It’s illustrated that Raman spectroscopy could be realized identification realgar fast and no damage.3. The research on acute toxicity and pharmacodynamics of realgar and its processed productsAccording to the differences of As2O3content and crystal structure of each sample, we selected five samples for the study, the samples including original realgar, acid processed, pieces which from Xinhehua and original realgar, acid processed which from Hunan yao fang.(1) The acute toxicity experiment show that the toxicity of processed products were lower than original realgar and the sample which has high content of As2O3will lead to high mortality rate. There had no significant difference on behavioral activity, organ characterization between surviving animals and blank animals.(2) To establish mouse with transplantation of S180tumor models, realgar was orally administrated at does of high, higher, medium, lower, low(8,4,2,1.0.5g·Kg-1) once a day for10d to observe tumor inhibition rate and the impact of immune organs on tumor-bearing mice. Filter out the right dose, then observe tumor inhibition rate and the impact of immune organs under five test samples. The results show that all realgar samples on S180bearing mice were inhibited, its rendered dose-dependent relationship. And to certain extent, the intensity of inhibitory effect was positively correlated the content of As2O3of each sample. |
