The coptis is rhizoma of Coptis chinensis Franch., Coptis deltoidea C.Y.Cheng et Hsiao, and Coptis teeta Wall, with Qingre dampness, Xiehuo detoxification function. At present, Research of Rhizoma Coptidis mainly concentrated in the alkaloids qualitative and quantitative, and pharmacological effects, however, there are few reports for the biological activity of polysaccharide and composition of research.Purposes:Polysaccharide from Rhizoma Coptidis of extraction method is studied, composition monosaccharide control methods have been established, and its polysaccharide and alkaloid component compared in vitro inhibiting a-glucosidase activity, cultivation and quality control to provide the scientific basis.Methods:1.Utilization Box-Behnken design-response surface methodology, the polysaccharide from Rhizoma Coptidis reflux extraction process four main factors to be assessed, these four factors are extraction temperature, extraction number, extraction time and water to material, as well as the their interactions, they affect three response value of extraction yield, polysaccharide yield and uronic acid yield.2.Application of1-phenyl-3-methyl-5-pyrazolone (PMP) pre-column derivative HPLC method, three Rhizoma Coptidis varieties composition monosaccharide analysis method has been established.3.Using the in vitro method of inhibiting a-glucosidase activity, its polysaccharide and alkaloid component compared in vitro inhibiting a-glucosidase activityResults:1. Extraction method Polysaccharide from Rhizoma Coptidis of major factor levels have been determined by single factor studied, as follows:extraction temperature of80℃to100℃, extraction time in2h to4h, extraction times1-3times, and water to material of1:10-1:20. Using Box-Benhnken designs,29experiments of selective combination were carried out. they affect three response value of extraction yield, polysaccharide yield and uronic acid yield, the polysaccharide from Rhizoma Coptidis final optimum extraction conditions is in the100℃extraction temperature, extraction time in3.8h, extracted3times, water to material of1:15.7.2. HPLC quantitative analysis method has been successfully established about different varieties of monosaccharide composition of Rhizoma Coptidis, having good repeatability and selectivity, the monosaccharides average recovery is97.09-99.14%, composition polysaccharide of three kinds is the same, mannose, C. chinensis’s rhamnose, glucuronic acid, galacturonic acid, glucose, galactose and arabinose from monosaccharide composition of the molar ratio of0.20:1:0.08:6.35:3.63:1.89:3.20, the C. deltoidea’s0.39:1:0.14:6.35:4.83:2.38:2.77, the C. teete’s0.38:1:0.09:5.83:5.70:2.44:4.40.3.Both alkaloids and polysaccharide from Rhizoma Coptidis parts have preferably inhibit a-glucosidase activity, which the alkaloid’s the IC50is171.67, polysaccharide is296.89. The two different ratios on the a-glucosidase inhibition have big difference, the ratio of polysaccharides and alkaloid is3:1, the IC50is the lowest. This ratio inhibition of a-glucosidase activity is the best.Conclusions:1.The optimum extraction conditions is at extraction temperature100℃, time3.8h, numbers3and ratio of water to material1:15.7.2.After hydrolysis of polysaccharides, these derived products have a strong UV absorption at250nm. The method has a good repeatability and stability,3.Both alkaloids and polysaccharide from Rhizoma Coptidis parts have preferably inhibit a-glucosidase activity, after the combination of the them is better than one part in inhibiting a-glucosidase activity.
