The extract is the most important intermediate of Chinese materia medica preparation. The complicated physico-chemical property usually had significant effect on the preparation and quality evaluation. Therefore, the evaluation mode of pre-preparation of TCM extract should reflected in the characteristic of Chinese materia medica preparation.The extract is the most important intermediate of Chinese materia medica preparation, the preparation and quality evaluation of which is significantly affected by the complicated physico-chemical property. Therefore, the evaluation mode of pre-preparation of TCM extract should reflected in the characteristic of Chinese materia medica preparation.In this study, Rhodiola extract was selected to be the object. Physical, chemical and biologic technologies were applied to study the nature of micromeritics, quality and biopharmaceutics of the exact. An evaluation mode, physics-chemistry-biology, which was applied in pre-preparation, was established in the study. A new route and method for dosage forms design, technology optimization, preparation craft optimizing, quality evaluation and rational clinical administration was suggested.The research of the micromeritics of Rhodiola extract showed it was common powder, the color was marron. D0.5was84.39μm and do.9was207.86μm. The exact was irregular mass under the microscope, with comparatively large angle of repose. The critical relative humidity was62%and the rate of hydrating was2.3μL·s-1. IR showed that mark absorption exited on3390,2933,1616,1516,1448,1367,1236,1038and579cm-1. DSC showed that a series of exothermic peak exited between100and140℃, and the peak was118.86℃. This essay identified the pharmaceutical nature of difference fluidity, strong hygroscopicity and short of hydrophilism.This essay determined the content of extractum, salidroside, tyrosol, polysaccharide and tannin in Rhodiola extract, and established a comparatively complete quality evaluation system of Rhodiola rosea extract. The research showed that the content of water soluble extractive was79.22%, the content of alcohol soluble extractive was79.61%, the content of salidroside was2.40%, the content of tyrosol was0.18%, the content of polysaccharide was15.86%, and the content of phenol was21.30%.(The content of tannin was13.81%.) The biopharmaceutics properties both in vivo and in vitro of the two pharmacodynamic components of Rhodiola extract was studied, with four level "dissolution in vitro-octanol/water partition coefficient-Intestinal Absorption in vivo-oral pharmacokinetics". Results showed that the dissolution rate of tyrosol in Rhodiola rosea extract was significant larger than salidroside in three medium medium.1g P of salidroside was from-1.22to-0.85, and1g P of tyrosol was from0.84to1.23. Salidroside was not absorbed in the small intestine of rats, however, tyrosol was well absorbed in the small intestine of rats. The kinetic parameter was, ka0.591±0.083h-1,10/21.19±0.15h, A%37.11±2.06%. Salidroside and tyrosol were rapidly absorbed and rapidly eliminated when they were intragastric administrated in rats. The absorption and elimination of tyrosol, high, middle and low dosage, were significant fast than salidroside. AUC of tyrosol was significant bigger than Salidroside.The project provided scientific accordance for the development of the preparation of Hongjingtian, meanwhile, it also provided a new research mode for the pre-preparation evaluation of intermediate of Chinese materia medica preparation.
