This paper proposes an immuno-affinity test column (IATC) to research the detection regarding the residual of Fumonisins B1(FB1)and Chloramphenicol (CAP) in food.In order to prepare Anti-FmB1gel and Anti-CAP gel, respectivel, ycouple Fumonisins antibody or Chloramphenicol antibody with Ago-Gel activated by hydrogen bromide, combine with the antibody of HRP to make their HRP gel, seal the Ago-Gel activated by hydrogen bromide to obtain the sealing gel. Antibody gel and HRP gel are mixed with sealing gel to achieve Detection layer and Control layer respectively and put into chromatography column, optimize and study the factors related the experience such as ionic strength and pH, establish a immune chromatography method with time-saving convenience, high sensitivity and visibility. It can detect Fumonisins B1and Chloramphenicol in food qualitatively and semiquantitatively.Concerning Fumonisins immune affinity column, it shows a great result of visual coloration when mixing ration of sealing gel and antibody gel is1:10. Sealing gel is mixed with Antibody gel which the ratio is1:90to obtain Control layer. The FB1-HRP with1:8000dilution is used for detection of Gel detection column. The limit of detection (LOD) of Fumonisins B1in the experience is40μg/L. Detect the Fumonisins residue in beer, white spirit, corn starch and Grain milk:dilute beer and white spirit directly for5times; put PBS into corn starch, extracted by the centrifugation method and then dilue it for5times; extracted the upper layer by the centrifugation method and dilute10times.Concerning Chloramphenicol immune affinity column, it shows a great result of visual coloration when mixing ration of sealing gel and antibody gel is1:20. Sealing gel is mixed with Antibody gel which the ratio is1:100to obtain Control layer. The CAP-HRP with1:200000dilution is used for detection of Gel detection column. The LOD of Chloramphenicol in the experience is1μg/L. Detect the Chloramphenicol residue in milk,milk powder, fish water and honey:detect milk and pond water directly; diluted milk powder for5times and dissolved it added PBS; dilute honey for5times.This method needs no organic solvent, complex pre-treatment and assist of large-scale instrument. It added standard of Fumonisins and Chloramphenicol of three concentration, is verified by the method of ELISA and obtained the same result which shows that the method has a great accuracy and can meet the requirements of rapid detection.
