Objective To study the expression of key molecules of the Wnt/β-catenin signalingpathway, such as Wnt2b,β-catenin,GSK-3β and CyclinD-1, analyze their correlation andtheir clinic pathology in gastric carcinoma tissue, and explore the relationship betweenWnt signal pathway and tumorigenesis, progression of gastric carcinoma.Methods48cases of gastric carcinoma and normal gastric mucous membrane, and24cases of paracancerous tissue were accumulated and made into one tissue microarrayswhich containing120dots. By tissue microarray-based NovolinkTMpolymer detectionimmunohistochemistry assay, a paralleled study was conducted to check the expression ofWnt2b, β-catenin, GSK-3β and CyclinD-1in gastric carcinoma, paracancerous tissue andnormal gastric mucous membrane. The relation between protein expressions andclinicopathological features in gastric carcinoma were analyzed and Spearman’scorrelation analysis was used to investigate the relationship among the four Proteinexpressions using SPSS17.0statistical software for Windows.Results1.It showed that the positive expression rate of Wnt2b,β-catenin,GSK-3β and CyclinD-1in GC were81.3%,66.7%,35.4%and45.8%, respectively.The positive expression rateof Wnt2b,β-catenin and CyclinD-1protein in gastric carcinoma was higher than that innormal tissues and paracancerous tissues and statistic difference is apparent byomparison (χ2=54.205,65.455,20.664,P<0.05).However, GSK-3β expression incarcinoma was lower than that in normal tissues and paracancerous tissues (χ2=26.968,P<0.05),there is statistic significance likewise.2.There was no correlation between the expression of Wnt2b,β-catenin,GSK-3β andCyclinD-1with gender,age of the patients and tumor size. The expression of Wnt2b wasclosely related to pTNM stage,cell ifferentiation degree and lymph node metastasis of GC(P<0.05). The expression of β-catenin was not only closely related to TNM stage,cell differentiation degree and lymph node metastasis, but to histological type,andnerve invasion of GC(P<0.05). The expression of GSK-3β was closely related to celldifferentiation degree, histological type and lymph node metastasis of GC(P<0.05).The expression of CyclinD-1was closely related to cell differentiation degreeand lymph node metastasis of GC(P<0.05).3.There was significantly positive correlation between the expression of Wnt2b andβ-catenin protein(r=0.340,P<0.05);There were negative correlations between theexpression of GSk-3β and Wnt2b、 β-catenin and CyclinD-1Protein inGC(r=-0.425,-0.493,-0.331,P<0.05), respectively; We also found that there were notsignificant correlation between the expression of CyclinD-1and Wnt2b, β-cateninprotein in GC(r=0.013,0.118, P>0.05), respectively.Conclusions1. The positive expression rate of Wnt2b,β-catenin and CyclinD-1protein in gastriccarcinoma was higher than that in normal tissues and paracancerous tissues. However,GSK-3β expression in gastric carcinoma was lower than that in normal tissues andparacancerous tissues.2. The expression of Wnt2b, β-catenin,GSK-3β and CyclinD-1protein in GC illustrate thatthe abnormal activation of Wnt/β-catenin signaling pathway play important role in thecarcinogenesis,invasion,metastasis and development of gastric carcinoma.3.There was positive correlation between the expression of Wnt2b with β-catenin protein.There were negative correlation between the expression of GSk-3β with Wnt2b,β-catenin and CyclinD-1protein in GC, respectively. It showed that the key moleculesof Wnt/β-catenin signaling pathway maybe interact with enhancement and synergisticeffect in the development and progression of GC carcinogenesis,promoting invasionand metastasis of GC together.
