Gastric carcinoma is one of the common malignant tumors, and is located the first place at incidence and death rate of all kinds of tumors in China. Occurrence of gastric carcinoma is a multiple factors,multiple steps process and involves the various mechanisms. At present, it is considered that pathogenesis of gastric carcinoma relates to Cx32,E-cad and Tau protein of gastric tumor cell.To investigate the relationship between Cx32, E-cad,Tau protein and clinical pothology characteristics(differentiation, metastasis of lymph nodes) of gastric carcinoma by detecting the expressions of them in normal and carcinomatous gastric tissue, for providing theoretical basis to the study of the pathogenesis of gastric carcinoma and new targets of clinical treatment.Part I The relationship between the change of gap junctions, gap junctions protein Cx32and gastric carcinomaObjectiveTo reveal the relationship between Cx32and gastric carcinoma through detecting the expression of Cx32in normal and carcinomatous gastric tissue by immunohistochemisty and RT-PCR, and also observing the change of gap junctions with transmission electron microscope.MethodsTo collect60cases of gastric carcinoma and10cases of normal gastric tissue(away the tumor more than10centimetres) which were surgically removed and certified by pathology in Chengde Medical College affiliated hospital from December,2008to March,2010. Immunohistochemisty was adopted to detect the expression of Cx32in normal and carcinomatous gastric tissue; to observe the difference of gap junctions in normal and carcinomatous gastric tissue with transmission electron microscope;the expression of Cx32mRNA was detected by RT-PCR in normal and carcinomatous gastric tissue.To analyse the experimental data combined clinical pothology with statistics.Results1. Immunohistochemisty1.1The expression of Cx32in carcinomatous gastric tissue was lower than that in normal gastric tissue (P<0.05).1.2The expression of Cx32in gastric carcinoma which were well differentiated and moderately differentiated was higher than that in poorly differentiated gastric carcinoma (P<0.05).1.3The expression of Cx32in gastric carcinoma without lymph node metastasis was lower than that in gastric carcinoma with lymph node metastasis (P<0.05).2. Transmission electron microscopeThe structures of gap junctions were damaged in gastric carcinoma.3. RT-PCR3.1The expression of Cx32mRNA in carcinomatous gastric tissue was lower than that in normal gastric tissue (P<0.05).3.2The expression of Cx32mRNA in gastric carcinoma which were well differentiated and moderately differentiated was higher than that in poorly differentiated gastric carcinoma (P<0.05).3.3The expression of Cx32mRNA in gastric carcinoma without lymph node metastasis was lower than that in gastric carcinoma with lymph node metastasis (P<0.05).Conclusions1. Immunohistochemical positive signals of Cx32were significant differences between normal gastric tissue and gastric carcinoma,also were concerned with differentiation degree and lymph node metastasis of gastric carcmoma.2. Cell gap junctions in normal gastric tissue were complete, but those in gastric carcinoma were damaged and incomplete.3. The correlation coefficient of Cx32and E-cad is:r=0.67(P<0.01);the correlation coefficient of Cx32and Tau protein is:r=-0.47(P<0.01);the correlation coefficient of E-cad and Tau protein is:r=-0.42(P<0.01).2. The correlation coefficient of Cx32mRNA and E-cad mRNA is: r=0.88(P<0.01);the correlation coefficient of Cx32mRNA and Tau mRNA is:r=-0.88(P<0.01);the correlation coefficient of E-cad mRNA and Tau mRNA is:r=-0.81(P<0.01).ConclusionsThe correlation of Cx32and E-cad was positive,the correlation of Cx32mRNA and E-cad mRNA was positive; The correlation of Cx32and Tau protein was negative,the correlation of Cx32mRNA and Tau mRNA was negative; The correlation of E-cad and Tau protein was negative,the correlation of E-cad mRNA and Tau mRNA was negative in gastric carcinoma.
