Objective:Based on the techniques of real-time PCR, the morphology of mice with FGIDs was observed. The impact of medicinal cake-separated moxibustion(MCSM) intervention was studied toward the amount of ACTH excreted by the hypophysis of the mice with FGIDs and GC from the adrenal gland caused by the chronic stress.Method::48SD mice were randomly divided into4groups with12ones in each group and the separate group was named as blank group (A), model group (B), model with MCSM group (C) and model with Xiaoyao San group (D). Except Group A, the other3groups were treated with molding (chronic restraint stress+overfatigue+indiscipline dieting, restraint was combined with clipping tails and shaking) for continuous21days. Group C was treated with MCSM for30mins in advance while the rest of the groups were tied up without moxibustion. Group D was perfused with Xiaoyao San while B and C with equivalent normal saline. All of the mice were fasted for24hours at the22nd day. After the fasting and anesthesia, the hypophysis, adrenal gland and gastric tissue of mice were dissected and observed by using the Real-time PCR and Electron Microscope.Results:1.After molding, obvious change were observed from the macro representation of all molded mice including drier and less shining hair, less activity, more instable motion, easier-to-be-angered and bowels either drier or more watery. But there was no specific change discovered in the blank group. In addition, the weight and appetite of the mice except those in the blank group was decreased evidently. 2. After molding, the excretion of ACTH and GC in the molding groups was improved with statistical significance (P<0.05or P<0.01). Compared with the molding group, the excretion of ACTH and GC was descended in MCSM group and Xiaoyao San group with statistical significance (P<0.05or P<0.01). But it should be emphasized that there was no significant difference.3. Compared with the blank group, the gastric residual rate was increased obviously in the molding groups, and the intestinal propulsion rate received evident decrease, which was proven with statistical significance(P<0.05or P<0.01). In conclusion, the gastric emptying time for the molded mice was prolonged while the intestinal propulsion function was impared. Compared with the molding group, the gastric residual rate in MCSM group and Xiaoyao San group was lowered clear while the intestinal propulsion rate was increased which was proven with statistical significance(P<0.05or P<0.01).4. For all of mice in four groups, the gastric mucous membrane was structure intact and cells were arranged in order with clear and continuous submucosa and muscular layer. Some molding group mice were observed with hemangiectasis for the inner circular muscle and inflammatory cell infiltration, which made fewer sense compared with the normal groups.Conclusion:MCSM is able to inhibit the expression of ACTH and GC in HPA axis. HPA axis is the important regulating access for curing the FGIDs with MCSM. It is possible to explain the mechanism of MCSM for curing the FGIDs since it can inhibit the expression of ACTH and GC.
