Objective:1. By study the effects of Dioscorea bulbifera L. used alone and combinated with Glycyrrhiza uralensis Fisch. on the biochemical indicator and oxidative stress indicator of the liver in rats to research whether Glycyrrhiza uralensis Fisch. can relieve the hepatic injury by inhibiting oxidative stress.2. By study the effects of Dioscorea bulbifera L. used alone and combinated with Glycyrrhiza uralensis Fisch. on the protein expression of CYP1A2and CYP2E1of the liver in rats to research whether Glycyrrhiza uralensis Fisch. can relieve the hepatic injury by inhibiting the protein expression of CYP1A2and CYP2E1.Methods:The SPF rats were randomly divided into six groups.After28days or35days of administration of drugs respectively,all rats were sacrificed to obtain the liver tissue:1. Serum glutamate pyruvate transaminase, glutamate oxalate transaminase and alkaline phosphatase were assayed.2. The contents of malondiadehyde and the activities of superoxide dismutase and glutathione peroxidase were surveryed.3. Chronic fluorosis rat liver tissues were taken to make histological section for observing the changes.4. The CYP1A2and CYP2E1protein expressions of the liver were determined by immunohistochemical method.Results:1. Compared with the control group,the levels of GPT,GOT,AKP were increased in the Dioscorea bulbifera L. Group(P<0.01).And35days of group is higner than28days of group.The levels of GPT,GOT,AKP were decreased in the combination group(.P<0.01or P<0.05or P>0.05).The effects of28days of the combination group was better.2. Compared with the control group,the contents of MDA were significamly increased(.P<0.01) and the activities of SOD and GSH-PX were significamly decreased in the Dioscorea bulbifera L. Group(p<0.01).And it associated with the time.However,the combination group degraded the contents of MDA (P<0.05or P>0.05).The activities of SOD and GSH-PX were increased(P<0.01).The effects of28days of the combination group was better.3. The liver tissue of rats with different grade pathologic change in the Dioscorea bulbifera L. group.And35days of treatment group was more obvious.The combination group can improve the liver tissue of rats with lesions.4. Compared with the control group,the protein expression of CYP1A2and CYP2E1of the liver in the Dioscorea bulbifera L. group were significantly increased (P<0.01).And35days of group is higner than28days of group.The protein expression of CYP1A2and CYP2E1of the liver in the combination group were decreased(P<0.05or P<0.01). The effects of28days of the combination group was better.Conclusion:Glycyrrhiza uralensis Fisch. can relieve the hepatic injury induced by Dioscorea bulbifera L. in rats by inhibiting oxidative stress and the protein expression of CYP1A2and CYP2E1. And it associated with the time.
