AIM:In this study, the effective part of Ledum palustre L. var. Angustum E. Busch were found by the research of cough expectorant and anti-inflammatory in mice. Analysis the effective chemical composition of Ledum palustre L. Provide a theoretical basis for the development of new drugs.METHOD:The Ledum palustre L. var. Angustum E. Busch ethanol extract obtained by heating reflux extraction with75%ethanol. The ethanol extract was extracted with petroleum ether, ethyl acetate and n-butanol respectively, and the petroleum ethe layer, ethyl acetate layer, n-butanol layer and aqueous layer were obtained. Ammonia induced cough model, intraperitoneal injection of phenol red and xylene coated ear swelling test were used to observe the effect’s of these parts.The layers were divided-into high and low dose groups administered sequentially, to determine the ledum palustre effective parts. The effective parts were used silica gel column chromatography, ODS column chromatography, preparative HPLC separation, and other modern technologies and methods to separate the components. Analysis the spectral data of these isolated compounds and identify its structure.RESULTS:1.Compared with other parts, the n-butanol layer and ethyl acetate layer can prolong mice cough incubation time induced by ammonia, and cough frequency was significantly reduced in3min, and can significantly promote the mice trachea phenol red excretion. Both of the n-butanol layer and ethyl acetate layer can significantly inhibited mice ear swelling.2.Ethyl acetate layer and n-butanol layer were separated,there are3compounds were separated and identified from ethyl acetate layer,wherein2were coumarin compounds and1were sterol compound. Therw are4compounds were separated and identified from n-butanol layer, these compounds were all flavonoids. CONCLUSION:The study determined the effective parts of cough expectorant and anti-inflammatory of Ledum palustre L. var.Angustum E.Busch,mainly in the n-butanol layer and the ethyl acetate layer.The main chemical components includ coumarin,sterols and flavonoids,namely:fraxin(fraxin), β-sitosterol (β-sitosterol),rutin(rutin),kaempferol3-(6"’-rhamnose-2"’-glucose glucosidase)(kaempferol3-(6"’-rhamnosyl一2"’-glucosyl一glucoside)), quercetin3-O-β—D—glucoside (quercetin-3-O-β-D-glucopyranoside), kaempferol-3-O-β-D-glucoside (Kaempferol-3-O-B-D-glucoside),6methoxy incense coumarin-7-β-D-glucoside (6-Methoxycoumarin-7-β-D-glucopyranoside).
