Objective Observe the effect of Banxia-shumi decoction on the depressive behavior and the monoamine neurotransmitters of depressive mice model, and throught it to probe the anti-depressive mechanism of Banxia-shumi decoction.Methods①Experimentation about the depressive behavior of depressive mice model:each time50ICR male mice were divided into5groups as follows:the blank control group, positive control group (clomipramine hydrochloride group), Banxia-shumi decoction groups(25.72、12.86、6.43g-kg-1), all groups were intragastrically administered once a day for10days. The tests of spontaneous motion, tail suspension (TST), forced swimming (FST) and antagonism of reserpine in mice were carried out to estimate the change of behaviors of depressive mice model and evaluate the antidepressant effect efficiency of Banxia-shumi decoction.②Detectation about the monoamine neurotransmitters of the mice brains:Chronic unpredictable stress are applied successively to set up depressive mice model. Enzyme linked immunosorbent assay (ELISA) is applied to detect the monoamine neurotransmitters (5-HT、DA and NE) of the brains.Results①Middle-dose and high-dose of Banxia-shumi decoction (12.86、25.72g-kg-1) shortened the dead time of tail suspension and forced swimming in mice(P<0.05or P<0.01). All doses of Banxia-shumi decoction had no effect on spontaneous motion (P>0.05) and resisted the descent of body temperature(P<0.05or P<0.01). Moreover, middle-dose of Banxia-shumi decoction resisted drooping eyelids (P<0.01)while high-dose of Banxia-shumi decoction also resisted the spasticity induced by the injection of reserpine after one hour(P<0.05or P<0.01).②Middle-dose of Banxia-shumi decoction promoted the cerebral NE concentration in the brain (P<0.05) while high-dose of Banxia-shumi decoction also promoted the cerebral5-HT concentration in the brain (P<0.05or P<0.01). All doses of Banxia-shumi decoction did not promote the cerebral DA concentration in the brain (P>0.05). Conclusion Banxia-shumi decoction has anti-depressant effect and can relieve the depressant behaviors of mice. The mechanism mainly is due to increasing the cerebral5-HT and NE concentration in the brains.
