| Objective:Metastatic ovarian cancer is a leading cause of cacer-related death in women.Cancer stem cells(CSCs) are involved in tumor progression and metastasis.And differentiation of cancer stem cells belongs to the capcity of self-renew.Like other oncogenes,overexpression and ectopic disperion of IQGAP1is related to the caner progession.To elucidate the link between IQGAP1and differentiation of ovarian cancer stem cells.Method:CSCs were enriched by serum-free suspension culture from3AO cell line,and medium with10%FBS was used to induce the differentiation of CSCs. Immunofluorescence was for dececting the stem markers,OCT4and SOX2.Realtime qRT-PCR and Western blotting were performed to determine the mRNA and protein expression of IQGAP1.The ability of cell migration and invasion was evaluated by scratch assay and Transwell chamber assay.The change in cell’s proliferation was detected by the wst-1kit.Results:1.CSCs,which were convenient enriched by serum-free suspension culture,expressed OCT4and SOX2.2.Comparing3A0cell line, CSCs seldom expressed IQGAP1.3.The ability of migration and wound healing were decreased after silencing IQGAP1except the proliferation ability..4.The differentiation decreased expression of OCT4and SOX2, and enhanced the invasion ability of CSCs.5. Silencing IQGAP1during CSCs differentiation could decrease the invasion ability.Conclusion:Free-serum culture is one of convenient assays for enriching CSCs. We investigated the increased IQGAP1could contribute to the aggressive behavior during differentiation of epithelial ovarian cancer stem cells. |
