Effects Of RhoC Silencing Combined With Rapamycin On The Expression Of Survivin Gene In Hepatocellular Carcinoma Cells

Objective:HCC is one of the most malignant tumor，And most patientshave been in later period when the symptoms are emerging.So this disease hasthe charactistics of low survival rate and high mortality rate. There are numerousreaserch results for the molecular events. The most studies of molecularmechanism are signaling pathways.The relevant signaling pathways about the proliferation, invasion,migrationand apoptosis are RhoC and PI3K/Akt/mTOR. This experiment which studiesthecombination of RhoC-siRNAwith Rapamycin influencing on theexpressi-onof Survivin in hepatocellular carcinoma cell BEL7402proves that thecom-bination could significantly inhibit the expression of Survivin andpromotetheapoptosis of HCC,this research also provides experimental evidencesfor explo-ringthe new remedy target for the HCC.Methods: This experiment was divided into six groups：Blank control7402group,negativecontrol Scramble-siRNA group, negative control Scramble-siRNA combined with Rapa group, Rapa group, RhoC-siRNAgroup,RhoC-siRNA combined with Rapa group. Western blot analysis was identifiedfor theeffe-ct of transfection of Survivin protein.The anti-apoptosis factorsurvivin was determined by PCR,and the expression of Survivin was detected by immunocyto-chemistry.Results: The expression of Survivin in different groups: negative controlScramble-siRNA with Rapa group,Rapa group andRhoC-siRNA groupcompared to blank control7402group and negative control Scramble-siRNAgroupare decreased(P<0.05), the expression of RhoC-siRNA combined withRapa group are significantly reducing (P<0.01), the Scramble-siRNA with Rapagroup, Rapa group and RhoC-siRNA group could inhibit the expression ofSurvivin (P<0.05), The RhoC-siRNA with Rapa group obviously inhibit theexpression of Survivin (P<0.01).Conclusions: The combination of RhoC-siRNA with Rapa could restrictevidently the expressionof Survivin,and this combination promote the apoptosisof HCC dramatically.
