Meanwhile, we transfected SW480cells with miR-1826inhibitor,investigated the effects of transfection on cell proliferation, cell cycle, apoptosis andinvasion in SW480cell line. Probe the biological role of miR-1826in colorectal cancer(CRC), lay the foundation for target gene therapy of CRC.Methods: We detected miR-1826expression levels by quantitative real‐timePCR(qRT-PCR) in72colorectal cancer tissues, their corresponding adjacent non‐tumortissues (NATS) and five colorectal cancer cell lines. The SW480cell line, which had thehighest elevation of miR-1826expression of the five cell lines tested, was transfected withmiRNA inhibitor that down-regulated the expression of miR-1826. Cell cycle andapoptosis were analyzed by using flow cytometry. Cell invasion was measured in transwellchambers and cell migration was was performed using wound‐healing method.Results: The expression of miR-1826was higher in colorectal cancer tissues than inNATS. Moreover, high expression of miR-1826was associated with indicators ofenhanced malignant potential, such as regional lymph node metastasis (p=0.018), advancedclinical stage(p=0.004) and poor prognosis (p=0.032). The SW480cell line, which had thehighest elevation of miR-1826expression of the five cell lines tested, was transfected withmiRNA inhibitor that down-regulated the expression of miR-1826. Successful transfectionwas confirmed by qRT-PCR. We also demonstrated that miR-1826inhibitor suppressedthe proliferation of colorectal cancer cells, promoted their apoptosis and induced G0/G1cell cycle arrest. In addition, the invasion and migration of the transfected tumor cells wereinhibited.Conclusion: All together, our findings suggest that miR-1826may play a critical role in the carcinogenesis of colorectal.
