Objective:to study the bone marrow mesenchymal stem cells (mesenchymal stem cells, MSC) affect A498kidney cancer nude mouse transplantation tumor. Methods: cultivate people kidney A498cell and MSC, use CM-Dil marker MSC and proliferation ability and activity of marker MSC cells after comparison, build A498kidney cancer nude mice subcutaneous transplantation tumor model, a total of36experimental nude mice, when the nude mouse transplantation tumor length to diameter of0.5-0.8cm, will be randomly divided into2groups:a group of injection CM-Dil marker MSC group (MSC), another group of injection of normal saline (NS control group), among them, each group has6is used only for lifetime observation.Since vaccination day1of the tumor, diet, defecation, activity, and the next day to observe nude mice tumor growth;In different times after the injection of MSC (3days,6days,15days,30days) measured with vernier caliper nude mice subcutaneous transplantation tumor of length to diameter, short diameter, and calculate the subcutaneous transplantation tumors had accumulated, record on the day of injection to at least the time of death, to calculate the average survival time, groups of nude mice survival time differences compare two groups of nude mice. Respectively in MSC injection after inoculation3days,6days,15days,30days, will each executed three MSC group and NS control group in nude mice, file.usd10.00MSC group of subcutaneous transplantation tumor tissue frozen section, under the fluorescence microscope, to observe the CM-Dil marker MSC at different time points in nude mice tumor tissue distribution; Wood grain-eosin (hematoxylin-esosin, HE) dyeing after HE pathological observation. Take the above time points in mice tumor tissue section, Twist gene and protein constituting expression was detected by RT-PCR and Western Blot. Results:CM-Dil marker MSC activity and proliferation and unlabelled MSC has no obvious change; MSC group nude mice subcutaneously transplanted tumor within the first week after injection of slow growth, compared with NS control group with significant difference (P<0.05).MSC in injection after15days, MSC group of subcutaneous transplantation tumor growth has slowed,15days and30days, two groups of nude mice tumor volume increase than before, but there was no statistically significant difference (P>0.05);The lifetime of nude mice after injection of MSC, nude mice extended; Fluorescent frozen section in the tumor tissue tracking nude mice CM-Dil marker MSC, fluorescence microscope observation to CM-Dil marker MSC under the green fluorescence excitation, the cell membrane is bright orange-red, cytoplasm and the nuclei are round dark space, with the passage of time, the fluorescence intensity gradually weakened, Twist the expression of genes and proteins in MSC Twist group was obviously less than NS group, the difference was statistically significant (P<0.05).Conclusion:bone marrow mesenchymal stem cell transplantation significantly inhibit A498kidney cancer nude mouse transplantation tumor growth, and delay receptors in nude mice survival time, heralding the bone marrow mesenchymal stem cells in the treatment of renal cancer between great potential and prospects; At the same time, the expression of Twist in the process showed a significant correlation, is expected to become the tumor biology behavior important indexes such as the occurrence and development, is likely to be between bone marrow mesenchymal stem cells with novel targets for the treatment of tumor.
