Abstract/Summary:
Objective: Liver fibrosis is often accompanied by iron deposition, irondeposition in liver fibrosis, and play an important role in the development ofliver fibrosis. Established animal models of liver fibrosis, and using MRI T2*techniques to explore the diagnostic value for iron deposition associatedliver fibrosis. And the correlation between iron deposition and liver fibrosisstage.As well as the value of MRI T2*imaging diagnosis of liver fibrosis.Method:1. To establish the rabbit of liver fibrosis model: pure CCl4was induced by intraperitoneally injection in rabbits. Pure CCl4low startingdose to a small increase in dose method, two times per week byintraperitoneally injection to induce the formation of liver fibrosis in rabbits.All of the rabbits were killed at12weekend. The livers were resected forgross and morphological observation with HE and Masson staining..2.control group, four rabbits by intraperitoneal injection of1ml/kg saline,twenty rabbits were divided into experimental groups A, B, C, D group, byintraperitoneal injection of a low dose of pure CCL4, a small increase in thedose of the method of inducing rabbit liver fibrosis. at6,8,10and12weeksafter the administration of48h, take A, B, C, D group experimental group andcontrol group one rabbits, respectively,3%pentobarbitone sodiumpentobarbital anesthesia, underwent liver MRI scan. GRE T2-Star Mapimaging specific parameters, TR43.50ms, TE1.98-12.23ms (8TEs),thickness of2.0mm, the pitch2.0mm, Field of View260mm, matrix256×256.24h within the scan liver tissue line drawn HE staining, Masson staining,Prussian blue staining and liver iron concentration was measured. The raw data load to the MRI post-processing workstation, using the software thatcomes with R2-STAR Functool2processed to generate T2*pseudocolormap, three ROI set methods were drawn in the same level, namely, thewhole liver ROI, great ROI, small ROI and then measured liver T2*values.Results:1. By intraperitoneal injection of a small dose of pure CCl4,experimental rabbits mortality rate was16.7%.2. In the experimental grouprabbits liver cells and macrophages seen varying degrees of dark bluegranules, along with the formation of fibrous septa septal main distribution.3. Along with the extension of the medication time, the LIC went up while theMRI T2*value tested by three different ROI methods went down. The T2*value tested by three ROI methods can effectively separate the normal, mildand moderate iron deposition. The MR T2*value was negatively correlatedwith the LIC and could contribute to the curved regression equation with goodfitting degree (P<0.05). Which liver fibrosis stage and more lagre area ROImeasurement of liver T2*values were better correlation..4. Hepatic fibrosishistopathology stage and the liver T2*values had the negative correlation.However, the T2*values could not separate stages of the fibrosis.Conclusions:1. By intraperitoneal injection of a small dose of pureCCl4successfully established rabbit model of hepatic fibrosis, andeffectively reduce animal mortality.2. The process of rabbit liver fibrosisassociated with mild to moderate hepatic iron deposition.3MR T2*technique can safely and rapid determination of the degree of hepatic irondeposition The aspiration biopsy might be conducted to improve the accuracywith it.4. The MR T2*values had the negative correlation with stages of thehepatic fibrosis. However, it failed to separate these stages. The MR T2*hadthe limited value in separating the stages of hepatic fibrosis.
