| Objective:To explore the analgesic effects of Zhishang cataplasm on inflamatory pain, and investigate the Zhishang cataplasm effects on the levels of capsaicin receptor in dorsal root ganglion in formalin injected rats.Methods:36SD rats were included in the experiment and were randomly divided into three groups.12SD rats were modeling group, which injected5%formalin0.1ml in the left plantar.12SD rats were treatment group, which used Zhishang cataplasm as topical medication one day before injected5%formalin0.1ml in the left plantar, and12SD rats were control group. To observed the pain behaviors after modeling, and use Dubuisson scoring method as a standard for comparison. About0.5CM cut at the left ankle on his left foot, take muscle tissue peeled about50mg and the segment of L3-6dorsal root ganglion have been taken from rats in a specific time.Enzyme-linked immunosorbent assay, Real-time PCR and western blot were used to detected the expression levels of capsaicin receptor in rats dorsal root ganglion.Results:1.The modeling group and treatment group were both shown typical biphasic nociceptive behavior reactions, but the credits in pain response were lower in the treatment group than in the modeling group (P<0.05). 2.Enzyme-linked immunosorbent assay blank adsorption group compared with model group, the model group than in the control group, the treatment group had treatment into prostaglandin E2levels lower than the model group, group comparison among the three groups (P<0.05), there are statistics significance, but the relative difference in the expression of three groups of three time points were not statistically significant.3.Realtime-PCR have shown that the expression of capsaicin receptor were higher in the modeling group than in the treatment group, and the expression of capsaicin receptor were higher in the treatment group than in the control group, the comparison among three groups had statistically significant difference (P<0.05). In each group, the expression levels of capsaicin receptor in three time points were no statistically significant differences (P>0.05).4. Western blot have shown that the expression of capsaicin receptor were higher in the modeling group than in the treatment group, and the expression of capsaicin receptor were higher in the treatment group than in the control group, the comparison among three groups had statistically significant difference (P<0.05). In each group, the expression levels of capsaicin receptor in three time points were no statistically significant differences (P>0.05).Conclusions:Zhishang cataplasm can relieve inflammatory pain in rats, and have some analgesic effects in rat inflammatory pain. Zhishang cataplasm can reduce the level of expression of rat formalin model of inflammatory pain localized muscle tissue prostaglandin E2. The rats in the dorsal root ganglia of capsaicin receptors were stably expressed, and capsaicin in the formalin model of inflammatory pain significantly increased the amount of receptor expression. Zhishang cataplasm can affect the expression levels of capsaicin receptor in dorsal root ganglion in formalin injected rats. |
