Objective:To observe the β-eleme reverse human lung adenocarcinoma cells PC9of gefitinibin-resistant in vitro. To explore the effect of β-elemene on resistant cells P-gp, p21, survivin protein expression in order to clear β-elemene reversal mechanism of action human to lung adenocarcinoma cells of gefitinib-resistant in vitro.Methods:MTT (MTT) colorimetric assay for PC9/ZD gefitinib resistance, resistance to multiple computing. MTT assay of non-cytotoxic concentrations of β-elemene and detect this concentration role reversal PC9/ZD calculated reversal fold. Western Blot detect the expression of resistance-associated protein P-gp, p21and survivin.Results:Gefitinib for PC9cells with an IC50of0.21μmol/L, for PC9/ZD cells with an IC50of5.45μmol/L,25.9multiple drug resistance. When p-elemene concentration of less than10μg/ml, for PC9and PC9/ZD inhibition were less than10%, no cytotoxicity. β-elemene10μg/ml reversible PC9/ZD to gefitinib resistance reversal multiple of2.68, while the intracellular levels of P-gp and expression of p21、survivin was significantly reduced.Conclusions:β-elemene has reversed PC9/ZD cells Nike gefitinib role in resistant cells may be related to reduced P-gp, survivin expression level. In addition to β-elemene whether as other targeted drugs and chemotherapy drugs reversal agent, pending further study.
